Purpose The aim of this study was to prepare fluconazole (FLC) nanoparticles coated with polyethylene glycol (PEG) in the form of FLC-PEG-NPs and optimize the size and entrapment efficiency. Methods Nine formulae were prepared by solvent antisolvent precipitation technique according to full 3 2 factorial designs. The effects of PEG molecular weight (X 1 ) and the drug polymer ratio (X 2 ) on the particle size (Y 1 ) and entrapment efficiency (Y 2 ) were explored. The prepared FLC-PEG-NPs were investigated for particle size, count rate, PDI, zeta potential, and morphology. Carbopol hydrogel was prepared, loaded with optimized FLC-PEG-NPs, and characterized for pH, FLC content, viscosity, homogeneity and spreadability, in vitro release, skin permeation, and antifungal activity. Results The formulated nanoparticles were uniform in size and spherical in shape with slightly rough surface and free from aggregations. The effect of PEG molecular was antagonistic on the particle size and was agonistic on EE %. The release of drug from hydrogel containing pure FLC was always lower than that from hydrogel containing FLC-PEG-NPs. The kinetic analysis of drug release obeys first-order release model and super case II transport mechanism. The cumulative amount of drug permeated applying hydrogel containing optimized FLC-PEG-NPs was significantly higher than the amount permeated using pure fluconazole containing hydrogel. The antifungal activity of hydrogel containing FLC in the form of optimized PEG-coated nanoparticles was better than hydrogel containing pure drug as indicated by relatively high inhibition zone using agar well-diffusion method. Conclusion Small spherical FLC nanoparticles with enhanced in vitro drug release as well as improved antifungal activity could be achieved by using PEG-coated fluconazole nanoparticles.

The present investigation aims at developing models by response surface methodology (FCCCD) followed by the scale-up method in preparing control release microsponge particles loaded with 5- fluorouracil, a drug used to treat actinic keratosis and colon cancer, and producing a new Dermal Delivery System. The polymer-based (ethyl cellulose and eudragit RS 100) microsponge particles were prepared by the w/o/w double emulsification method. The optimized product was formed with the combination of independent variables levels: polymer (600 mg), stirring speed (1198 rpm) and surfactant (2% w/v), yielding responses as yield (~63.6257%), the average size of particles (~151.563 µm), entrapment efficiency (~75.319 %) and drug release in 8hr (~75.75%), with desirability value of 0.737. The products showed similar responses as obtained in scale-up work. FT-IR, DSC and SEM studies confirmed the drug's compatibility with polymers and porous morphology. Finally, gel embedded optimised product showed shear-thinning rheological property, ideal for drug release from the thixotropic gel.

The aim of the present work was to develop a microsponge delivery system of baclofen to control its release and thereby reducing dosing frequency and enhancing patient compliance. The microsponge was produced by oil in oil emulsion solvent diffusion method. The effects of drug/polymer ratio, stirring time and type of Eudragit polymer on the physical characteristics of microsponges were investigated. The prepared microsponges was characterized for production yield (PY), loading efficiency (LD), particle size, surface morphology, and in vitro drug release. The results showed that the microsponge formula with Eudragit RS100 had optimum physical properties with PY % equal to 97 %, and LD % equal to 81% and controlled drug release (75% of drug release in 8 hours) when compared with other formulas and pure BFN. Therefore, the non-‎aqueous emulsion solvent diffusion method is a promising method to produce baclofen microsponges.‎
