A novel mutation in PSEN1 (p.Arg41Ser) in an Argentinian woman with early onset Parkinsonism

Abstract: Mutations in presenilin-1 (PSEN1) account for the majority of cases of familial autosomal dominant early-onset Alzheimer's disease (AD) as well as in sporadic forms. Atypical presentations are reported including extrapyramidal signs. In the last years, a pleiotropic effect of some PSEN1 variants has been reported in Parkinson's disease (PD).

“…PS2 Gly34Ser was not detected in any individual without AD among the family members. Interestingly, PS1 Arg41Ser was identified in patients with early-onset PD [ 33 ]. Pathogenic PSEN mutations were mostly found in transmembrane regions.…”

“…Besides their established role in mediating the formation of Aβ peptide, more recently mutant PS1 has been shown to impair numerous cellular functions, such as calcium flux, organization of proteins in different compartments, and protein turnover via vacuolar metabolism 60 . Interestingly, a novel PSEN1 mutation was recently identified as the likely cause for early‐onset parkinsonism 61 …”

“…Parkinsonism is part of the PSEN1-associated phenotype; however, the occurrence of parkinsonism as an early sign is uncommon. We reviewed six reports (including our study) of seven patients with PSEN1 mutations and a disease onset symptom of EOP (Table 1) [3,[8][9][10][11]. The age at disease onset ranged from 25 to 42 years.…”
