A novel nucleocytoplasmic shuttling sequence of DAZAP1, a testis-abundant RNA-binding protein

Abstract: Deleted in Azoospermia Associated Protein 1 (DAZAP1) is a ubiquitous RNA-binding protein highly expressed in the human and the mouse testes. It shows a dynamic subcellular localization during spermatogenesis, present predominantly in the nuclei of late-stage spermatocytes and round spermatids and translocated to the cytoplasm during spermatid elongation. To test the hypothesis that DAZAP1 shuttles between the nucleus and the cytoplasm, we studied the nuclear transport of DAZAP1 in somatic cells using immunosta… Show more

“…The cells were stained with the anti-USP26-M antibody 17 and an Alexa Fluor 488 conjugated goat anti-rabbit IgG secondary antibody (Invitrogen, Carlsbad, CA, USA) according to Lin and Yen. 19 The acrosome was labeled with lectin peanut agglutinin (PNA) conjugated with Alexa Fluor 594 (Invitrogen) or a mouse monoclonal anti-acrosin antibody (GeneTex, Inc., Irvine, CA, USA), and the nucleus was stained with Hoechst 33258 (Invitrogen). The cells were then mounted and photographed with a Zeiss LSM510 confocal microscope (Carl Zeiss Microscopy GmbH, Jena, Germany).…”

“…Human DAZAP-1 is a nucleocytoplasmic shuttling protein (Lin & Yen 2006) and its X. laevis ortholog, PRRP, associates with mRNA localization elements in Vg1 and VegT mRNAs (Zhao et al 2001). However, it is unclear whether DAZAP-1 plays an active role in mRNA localization and its role on these mRNAs appears to be independent of xDAZL.…”

“…It is expressed must abundantly in the testis and to a lesser degree in other tissues (Tsui et al 2000;Dai et al 2001;Pan et al 2005). In somatic cells, DAZAP1 is present mainly in the nucleus, and its association with the hnRNP particles makes it a bona fide hnRNP protein (Lin and Yen 2006). In mouse testes, DAZAP1 shows a dynamic distribution during spermatogenesis.…”

“…A role of DAZAP1 in transcription is suggested by its specific exclusion from the transcriptionally inert XY body in the nuclei of pachytene spermatocytes, and the requirement of active transcription for its nuclear localization (Vera et al 2002;Lin and Yen 2006). Its ability to shuttle between the nucleus and the cytoplasm supports its role in mRNA transport (Lin and Yen 2006).…”
