A polymorphism in the 3’ untranslated region of Hypoxia-Inducible Factor-1 alpha confers an increased risk of cervical cancer in a Chinese population

Abstract: Summary. -In this study, a major part of genome of the pestivirus isolate 297 from Slovakia, comprising the 7195 nt-long 5΄-UTR-NS3 region was sequenced and analyzed. Conserved cleavage sites between individual viral proteins of this region were determined and the number of amino acids of respective proteins was estimated as follows: 168 for N pro , 100 for C, 227 for E rns , 195 for E1, 373 for E2, 70 for p7, 453 for NS2, and 683 for NS3. Based on sequence and phylogenetic analysis of 5΄-UTR, N pro , and E2 t… Show more

“…Our functional assay indicated the potential impact of rs2057482 on the post-transcriptional regulation of HIF1A gene by miR-199a-5p. Consistently, SL Fu et al 34 have also evaluated the HIF1A mRNA expression in 17 cervical cancer tissues with genotype data of SNP rs2057482 and found similar result, indicating that the tissues carrying CT/TT genotypes of rs2057482 had significantly decreased HIF1A mRNA expression levels when compared to those with CC genotype. Furthermore, we found that the significant or borderline significant protective effects of CT/TT genotypes of rs2057482 on OS and RFS were almost only observed in adverse strata patients, but not in favorable strata patients.…”

“…To the best of our knowledge, this is the first study to report that HIF1A gene polymorphisms may serve as an independent biomarker to predict the protective treatment response of adjuvant Several studies have performed case-control study to investigate the association between SNP rs2057482 in HIF1A gene and cancers. For example, a case-control study with 518 cervical cancer patients has found that a significantly increased risk of cervical cancer is associated with the CC genotype of rs2057482, and carriers of CT/ TT genotypes have significantly decreased HIF1A mRNA expression levels compared to those with CC genotype (Fu et al, 2014). In another case-control study, they have observed the significant associations of SNP rs2057482 with risk of rectal cancer (Frank et al, 2010).…”

“…In contrast, the association results of SNP 1772 C > T of HIF-1α gene with different kinds of cancers were not consistent in literature review. For example, the SNP 1772 C > T of HIF-1α gene was detected in several cancers [ 18 - 21 , 23 ] but it was absent for colorectal [ 34 ], and cervical [ 35 ] cancers.…”
