A quantitative and molecular examination of Tuber melanosporum mycorrhizae in Quercus ilex seedlings from different suppliers in Spain

Abstract: Aim of study: The aim of the work was to determine the degree of mycorrhization of Quercus ilex L. subsp. ballota (Desf.) Samp. by the black truffle fungus T. melanosporum Vittad. by quantitative and molecular analyses.Area of study: seedlings inoculated by different Spanish suppliers.Material and methods: The internal transcribed spacers (ITS) of mycorrhizae from different plants were amplified by nested PCR involving fluorescently-labelled primers, and the amplicons either directly sized by ARISA or analysed… Show more

“…Several ways to improve truffle production have been studied: management practices at the greenhouse stage [ 18 , 19 ]; standardized procedures to evaluate seedling quality for customers benefit [ 10 , 20 , 21 , 22 ]; and growth conditions promoting T. melanosporum proliferation in the field [ 23 , 24 , 25 , 26 ].…”

“…Reliable , simple techniques like the use of species-specific PCR primers for selective amplifications are recommended (Rubini et al, 1998;Mello et al, 2002;Benucci et al, 2011). We discourage laborious procedures like cloning previously amplif ied products and sequencing, as required by other methods of truffle-inoculated seedling certification (Alvarado and Manjon, 2013).…”

“…truffle-inoculated seedlings based on both morphological and molecular analyses (Govi et al, 1995;Fischer and Colinas, 1996;Reyna et al, 2002;Miko and GaÏo, 2007;Bach et al, 2010;Alvarado and Manjon, 2013). Each of these methods has prioritized specific issues, making them all different from each other.…”
