A randomized, double blind, placebo-controlled pilot trial of the safety and efficacy of atorvastatin in children with elevated low-density lipoprotein cholesterol (LDL-C) and type 1 diabetes

Abstract: Atorvastatin lowered LDL-C, apoB, and atherogenic lipoprotein subparticles in children with T1D and elevated LDL-C without worsening insulin resistance. The drug was well tolerated and safe. Long-term studies would provide better insight on the impact of these interventions in the development of cardiovascular disease in children with diabetes.

“…The overall decline in LDL-C and TC was somewhat less than that seen in children with familial hypercholesterolemia 15,16 but similar to that in children with Kawasaki disease 18 , Type 1 diabetes 26 and systemic lupus 17 . LDL-C and TC decreased by a mean of 39% and 31%, respectively, between baseline and 26 weeks in the 140 children randomized to atorvastatin in a placebo-controlled trial in children and adolescents with familial or severe hypercholesterolemia 15 .…”

“…Recently, a small randomized, double blind, placebo-controlled pilot trial that included 42 adolescents with T1DM and elevated low-density lipoprotein (LDL) cholesterol showed a decreased lipid levels by atorvastatin therapy. Atorvastatin was safe and well tolerated in this study (18). However, very few studies investigated the effect of statins on CVD outcomes.…”

“…As for the safety of statins in children, they have been shown to be safe and well-tolerated in children with hypercholesterolemia as well as those with diabetes [56,57]. In the pravastatin study previously described, the treatment was well-tolerated [55].…”
