A reactive oxygen species scoring system predicts cisplatin sensitivity and prognosis in ovarian cancer patients

Abstract: BackgroundTo reveal roles of reactive oxygen species (ROS) status in chemotherapy resistance and to develop a ROS scoring system for prognosis prediction in ovarian cancer.MethodsWe tested the sensitizing effects of ROS elevating drugs to cisplatin (cDDP) in ovarian cancer both in vitro and in vivo. A ROS scoring system was developed using The Cancer Genome Atlas (TCGA) database of ovarian cancer. The associations between ROS scores and overall survival (OS) were analyzed in TCGA, Tothill dataset, and our in-h… Show more

“…Prediction of treatment responses makes sense for HGSOC [10]. For HGSOC patients, treatment regimens are scheduled based on platinum sensitivity [37,38]. However, nearly 20% of patients are inherently resistant to platinum agents [1].…”

“…Moreover, improper platinum dosage is toxic and may import resistance to other drugs [3,4]. Some gene profile-based techniques combined with sequencing, microarrays, and PCR were developed to predict prognosis and drug responsiveness of ovarian cancer, but their application had limitations [38,39]. Moreover, detection of protein expression can be more effective than gene screening.…”

“…In this system, there are 25 scores in which scores 0-12 demonstrate low score groups, while scores 13-25 show high score groups. As ROS overgeneration enhances CP sensitivity and apoptosis induction, by enhancing ROS levels, patients are included in high score groups, which have high overall survival and good prognosis [102]. This score can be used in clinical course.…”

“…Accurate reactive oxygen species (ROS) quantification and characterization plays an important role in the investigation of metabolism [1,2], inflammatory responses [3,4], the pathogenesis of the disease [5,6], treatment monitoring [7][8][9] [10] and potentially provides prognostic information [11]. Currently, standard ROS measurement uses various molecular indicators such as dichlorofluorescein, a fluorescent probe that reacts intracellularly with peroxidase, or CellROX, a fluorogenic probe for measuring oxidative stress in live cells and tissue [12][13][14].…”
