“…This failure to detect a gB-specific CTL response in HSV-l-immune mice is in disagreement with the studies of Blacklaws et al (1987) and McLaughlin-Taylor et al (1988). The former authors demonstrated a minor population of HSV-l-specific CTLs from CBA mice (H-2 k) which lysed autologous L cells expressing the HSV-1 KOS gB gene, whereas McLaughlin-Taylor et al (1988) reported a population of CTLs specific for the HSV-1 F gB molecule demonstrable in BALB/c (H-2 d) mice. We addressed these discrepancies by using the combined techniques of limiting dilution culture and split well analysis to determine the recognition of VgB11-infected cells by HSV-l-specific CTLs at the clonal level.…”

“…HSV-I strain F (obtained from P. G. Spear, Northwestern University, Chicago, II1., U.S.A.) was propagated and titrated on Vero cell monolayers. Recombinant vaccinia viruses vgBll, which expresses HSV-I gB (McLaughlin-Taylor et aL, 1988) and VSC8, which expresses flgalactosidase (Chakrabarti et al, 1985), were obtained from Bernard Moss (National Institute of Allergy and Infectious Diseases, Bethesda, Md., U.S.A.) and were propagated on CV1 cells.…”

“…The observed differences in lysis of two H-2 b target cell lines each infected with AdgB2 highlights another of the potential difficulties in comparing CTL data where different cell lines have been used. MC57 cells infected with a recombinant vaccinia virus, vgB11, expressing HSV-1 gB (McLaughlin-Taylor et al, 1988) were also efficiently lysed by HSVspecific CTLs (Fig. 4).…”

“…In the case of herpes simplex virus type 1 (HSV-1), the identity of target antigens is unresolved and controversial. For example, of the seven major glycoproteins, three have been the subject of both positive and negative findings (Glorioso et al, 1985;Blacklaws et al, 1987;Rosenthal et al, 1987;McLaughlin-Taylor et al, 1988;Martin et al, 1989). The confusion appears partly attributable to the influence of the major histocompatibility complex (MHC) class I genotype, but the particular system used to express the viral genes may also affect the outcome (Coupar et al, 1986;D.…”
