A regulatory role for macrophage class A scavenger receptors in TLR4‐mediated LPS responses

Abstract: Recognition of microbial components by TLR, key sensors of infection, leads to induction of inflammatory responses. We found that, in vivo, TLR4 engagement by LPS induces upregulation of the class A scavenger receptors (SR) macrophage receptor with a collagenous structure (MARCO) and SR-A, which occurs, at least in the case of MARCO, via both MyD88-dependent and -independent pathways. When challenging mice with a low dose of LPS followed by a high dose, class A SR-deficient mice showed a higher survival rate t… Show more

“…The mechanism of NF-kB upregulation by LPS has been extensively researched. It is becoming clear that activation of TLR4 and SRA on the cell surface is involved in LPS-induced NF-kB activation.NF-kB activation and TLR4 mediate the secretion of LPS-induced inflammatory factors via a MyD88-dependent pathway [2]. And SRA may be involved in this process, particularly in the context of atherosclerosis [10].…”

“…SRA mediates the regulation of the B cell activation state [2] and suppresses CD8 T cell activation by down-regulating TLR4 signaling [3]. Evidence suggests that LPS regulates SRA expression in macrophages.…”

“…MARCO is known to be upregulated by bacterial Ag (such as LPS) (17) and, thus, was studied in the context of host defense against infections (18). Because it is known to be connected to downstream signals through TLR (19,20) we further assessed the effect of MARCO cross-linking in MyD88-, TLR2-, TLR4-, or TLR9-deficient mice. The data showed similar reductions in CD21 expression levels after anti-MARCO Ab treatment in all mice (Supplemental Fig.…”

“…Macrophages of mice have been shown to upregulate Marco via LPS-induced TLR4 signalling, which can be a mechanism to increase phagocytic activity (Chen et al, 2010). Similarly, when we infect zebrafish with S. typhimurium, an LPScontaining pathogen, we observe induction of marco (Kanwal et al, 2013).…”
