A restricted cell population propagates glioblastoma growth after chemotherapy

Abstract: Glioblastoma multiforme (GBM) is the most common primary malignant brain tumor, with a median survival of about one year1. This poor prognosis is due to therapeutic resistance and tumor recurrence following surgical removal. Precisely how recurrence occurs is unknown. Using a genetically-engineered mouse model of glioma, we identify a subset of endogenous tumor cells that are the source of new tumor cells after the drug, temozolomide (TMZ), is administered to transiently arrest tumor growth. A Nestin-ΔTK-IRES-… Show more

“…The prognosis of astrocytoma differs according to features such as age, disease stage, and histological type. GBM is the most malignant astrocytoma, with a median overall survival (OS) of approximately 12 months and a 5‐year OS of 4.8%‐5.4%,1, 2, 3 while patients suffering from anaplastic astrocytoma have a median OS of 38 months and a 5‐year OS of 25.9%‐41.1% 1, 4. A deeper understanding of the prognostic factors of GBM may provide new ideas for the prevention and management of this disease.…”

“…A minor cell population, constituted of multipotent tumor-initiating stem-like cells (GSCs), has been recently implicated in GBM recurrence and resistance to conventional treatments [7][8][9].…”

“…These confusing data are particularly troublesome in the case of the esophagus and the common premalignant lesion Barrett's Esophagus (BE). First there have been reports of fast cycling cells with stem cell capacity in animal models and some of them indicate a residual stem cell population is needed while others do not [11][12][13][14][15] . Second some additional reports have implicated endodermal (including epithelial) embryonic markers in man but their relevance to human stem cells remains unclear 16 .…”
