A restricted period for formation of outer subventricular zone defined by Cdh1 and Trnp1 levels

Abstract: The outer subventricular zone (OSVZ) is a germinal layer playing key roles in the development of the neocortex, with particular relevance in gyrencephalic species such as human and ferret, where it contains abundant basal radial glia cells (bRGCs) that promote cortical expansion. Here we identify a brief period in ferret embryonic development when apical RGCs generate a burst of bRGCs that become founders of the OSVZ. After this period, bRGCs in the OSVZ proliferate and self-renew exclusively locally, thereby … Show more

“…There are also genes that are differentially expressed with respect to the gyrus formation reported recently (e.g., Trnp1, Cdh1) (De Juan Romero et al 2015;Martinez-Martinez et al 2016). This guinea pig system would be useful for functional evaluation of those factors.…”

“…In contrast, the KO cortex contained overabundant Ki67+ NPC in the basal SVZ and intermediate zone (IZ) (Fig. 2e, f), reminiscent of the positioning of ORG that normally populate the OSVZ 1214,20 . Discontinuous clusters of basal NPC were accompanied by thinning of the VZ, suggesting that precocious OSVZ progenitors are derived by premature withdrawal from the VZ (Fig.…”

“…Studies in the ferret suggest there may be a restricted period for the formation of the oSVZ during which vRG cells generate large numbers of oRG cells. This window is delineated by down regulation of Trnp1 and Cdh1 [31]. The DNA-associated protein Trnp1 had previously been identified as a regulator of mammalian brain development, and shRNA knockdown in the mouse leads to increased production of oRG-like cells [32 •• ].…”
