A Role for PGC-1a in the Control of Abnormal Mitochondrial Dynamics in Alzheimer’s Disease

Abstract: Emerging evidence suggests that the proper control of mitochondrial dynamics provides a window for therapeutic intervention for Alzheimer’s disease (AD) progression. The transcriptional coactivator peroxisome proliferator activated receptor gamma coactivator 1 (PGC-1a) has been shown to regulate mitochondrial biogenesis in neurons. Thus far, the roles of PGC-1a in Alzheimer’s disease and its potential value for restoring mitochondrial dysfunction remain largely unknown. In the present study, we explored the im… Show more

“…Additionally, the relationship between mitochondrial biogenesis and fusion-fission dynamics is essential in maintaining mitochondrial functionality, and PPARGC1A promotes mitochondrial fusion-related proteins expression to formation of elongated, interconnected mitochondria. 47 In our research, ZEN treatment impaired mitochondrial biogenesis, excessive mitochondrial fission, accompanied with mitochondrial morphological abnormalities in goat EECs and ESCs. In general, mitochondrial biogenesis, fusion-fission dynamics, UPR mt , and mitophagy each have distinct responsibilities, but they are interconnected.…”

“…AMPK could activate Sirt1 by elevating the nicotinamide adenine dinucleotide (NAD + ) levels in cells, resulting in the activation of PGC1α ( Canto et al, 2009 ; Fulco & Sartorelli, 2008 ). AAV-mediated overexpression of PGC1α in the lateral parietal association reversed the damage and swelling of mitochondria in cortical neurons through improving abnormal mitochondrial dynamics ( Wang et al, 2022a ). Dong et al have defined that Aβ 25−35 inhibited the mitochondrial biogenesis in cultured primary hippocampal neurons via attenuating the AMPK/Sirt1/PGC1α signaling pathway ( Dong et al, 2016 ).…”

“…To overexpress FoxG1 in the hippocampus, the FoxG1 fl/fl males were crossed with Cre ERT females to achieve Cre/FoxG1 fl/fl double transgenic mice, and TM was administered by intraperitoneal injection to Cre/FoxG1 fl/fl mice. TM was dissolved in corn oil at a concentration of 20 mg/mL, and the dose administered was 75 mg/kg body weight once a day and sacrificed at day 14 to detect the changes in FoxG1 expression in hippocampus (Wang et al 2022a , c ).…”
