A rust fungal effector binds plant DNA and modulates transcription

Abstract: The basidiomycete Melampsora larici-populina causes poplar rust disease by invading leaf tissues and secreting effector proteins through specialized infection structures known as haustoria. The mechanisms by which rust effectors promote pathogen virulence are poorly understood. The present study characterized Mlp124478, a candidate effector of M. larici-populina. We used the models Arabidopsis thaliana and Nicotiana benthamiana to investigate the function of Mlp124478 in plant cells. We established that Mlp124… Show more

“…The exclusive GO categories related to the selected variants have already been reported to be associated with resistance. Sphingolipid metabolism is intimately connected to programmed cell death [123][124][125] ; DNA topological change is a wider category with different implications in many biological processes, including responses to pathogens 126 ; differences in nitrogen compound transport might be related to the accumulation of this nutrient and its influence on resistance against pathogens 127 ; and phosphatidylinositol-mediated signaling includes important categories that also act on plants' responses to pathogens 125 . A considerable number of metabolic pathways related to both BACs and the selected scaffolds were also detected.…”

“…During infection, plant pathogenic microbes deliver virulence proteins, known as effectors, into host cells to overcome plant immunity and promote parasitic colonization through the manipulation of cellular processes [1]. Once inside host tissues, effectors traffic to various cellular compartments where they interact with host proteins or nucleic acids and exert their virulence function [2][3][4][5][6]. To target these destinations, effectors possess domains or motifs in their sequence; for example, nucleus localized effectors can contain nuclear-localized signals (NLS) and some chloroplast localized effectors may carry a transit peptide [4,7,8].…”
