A Salmonella Small Non-Coding RNA Facilitates Bacterial Invasion and Intracellular Replication by Modulating the Expression of Virulence Factors

Abstract: Small non-coding RNAs (sRNAs) that act as regulators of gene expression have been identified in all kingdoms of life, including microRNA (miRNA) and small interfering RNA (siRNA) in eukaryotic cells. Numerous sRNAs identified in Salmonella are encoded by genes located at Salmonella pathogenicity islands (SPIs) that are commonly found in pathogenic strains. Whether these sRNAs are important for Salmonella pathogenesis and virulence in animals has not been reported. In this study, we provide the first direct evi… Show more

“…For instance, the FUR-regulated sRNAs RyhB and IsrE have been found to be highly induced in a fibroblast model of Salmonella infection (Ortega et al, 2012). Many pathogenicity island-encoded sRNAs (Isr genes) are activated inside macrophages (Gong et al, 2011;Padalon-Brauch et al, 2008) and numerous additional sRNAs undergo strong expression changes during growth over a range of infection-mimicking in vitro conditions (Kroger et al, 2013). Moreover, in vitro studies with Salmonella and pathogenic E. coli suggest various sRNAs that contribute to pathogenicity.…”

“…The Mlc global regulator has been shown to downregulate an hilE promoter (27). The small noncoding RNA isrM targets the hilE transcript to reduce the repressing activity of hilE (57). The LysR-type regulator LeuO has been shown to activate hilE transcription to repress HilD activity (58).…”

“…InvR sRNA acts as a repressor of OmpD protein synthesis (Pfeiffer et al, 2007). IsrM sRNA, which is encoded by a Salmonella pathogenicity island, targets the mRNAs coding for SopA, a SPI-1 effector, and HilE, a global regulator of the expression of SPI-1 proteins (Gong et al, 2011). SgrS sRNA post-transcriptionally represses the expression of the secreted virulence factor SopD .…”
