Recent technological advances have resulted in a dramatic renaissance of population genetics and its application to species ecology and conservation. This review summarizes the progress made in applying these new techniques, notably hypervariable genetic markers (microsatellite loci and mitochondrial control region), to the study of marsupials. Since 1990, population genetic studies have overwhelmingly been of marsupial species from eastern and southern Australia, largely focusing on threatened species or those with restricted distributions. To date, over 500 polymorphic microsatellite loci have been isolated from 38 marsupial species (six American, 32 Australian), including representatives from 13 of the 18 extant marsupial families. Levels of microsatellite diversity identified within the 209 marsupial populations (43 species) so far examined have varied greatly, although the range is similar to those reported from eutherians and other vertebrates. Marsupial populations with high levels of genetic diversity tend to be those from relatively abundant or widespread species, while those with lower levels are typically species with restricted distributions, that are threatened, found on islands or have been established via translocation. Although data for most families are still limited, bandicoots, koalas and wombats appear less and phalangerids more diverse than the marsupial average. The application of these hypervariable genetic markers to investigate marsupial species ecology has substantially improved our understanding of population biology, behaviour and reproduction in many species, as well as informing conservation initiatives and management plans for many threatened marsupial taxa. It is hoped that in the future, marsupial population genetic studies can be expanded to include a larger number of South American and New Guinean species, as well as a better representation of arid, tropical, widespread and abundant Australian species.

A new population of eastern short eared rock-wallaby (Petrogale wilkinsi) has been recorded in Queensland. The animals were photographed on a sandstone cliff face in Lawn Hill Gorge, Boodjamulla National Park, ~160km from the closest record on Wollogorang Station in the Northern Territory. The species was originally thought to occur north of the 600-mm rainfall isohyet, with most of the population found in the monsoonal tropics of the Top End. This record significantly extends the known range of this taxon to the south-east and places P. wilkinsi at the 450-mm rainfall isohyet. Sympatry in rock-wallabies is unusual but P. wilkinsi at Boodjamulla appears to be sympatric with the more common purple necked rock-wallaby (Petrogale purpureicollis). A recent taxonomic revision of the brachyotis group found that there was a significant mitochondrial DNA (mtDNA) divergence between the Wollogorang record and the rest of the P. wilkinsi group further north, but further genetic sampling across the southern Gulf of Carpentaria is required to determine whether these populations represent an additional distinct taxon.

Four morphospecies of Cloacina, parasitic nematodes in the stomachs of rock wallabies (Petrogale spp.) from Queensland, were compared genetically using sequence data of the two internal transcribed spacers of nuclear ribosomal DNA (rDNA). The results suggest that two geographically isolated populations of C. ernabella from P. purpureicollis were genetically distinct. Based on the autapomorphic species concept, these two C. ernabella populations represented different species. For the three other nematode morphospecies, there were genetic differences among individuals of a morphospecies present in different species of host. The results suggest that each may represent a complex of sibling species, with a different species present in each species of rock wallaby examined for that morphospecies. In the C. caenis and C. pearsoni complexes, the lineage present in P. purpureicollis from western Queensland represents a sister taxon to those in the P. pencillata complex from the east coast. In the C. robertsi complex, the taxon parasitic in P. persephone represents the sister taxon to those in the P. pencillata complex and in P. purpureicollis. C. robertsi was found for the first time in P. purpureicollis from Winton in central Queensland, suggesting contact in the recent past between populations of P. purpureicollis and a member of the P. penicillata complex.
