The skin plays a fundamental role in regulating the body’s internal balance and protecting against external traumas. A broad variety of environmental risk factors frequently result in acute skin wounds, whose inappropriate treatments would lead to chronic skin wounds that are difficult to heal. Traditional dressings have been widely used to repair chronic skin wounds, however their drawbacks such as insufficient hemostatic efficacy and non-moist environment have severely limited their clinical applications. As the principal component of skin, collagen has always been a research hotspot in the field of chronic skin wounds due to its advantages of low antigenicity, high biocompatibility and superior bioactivity. Collagen-based dressings have been increasingly developed to heal the chronic wounds during the past decades, arising from their capability in decreasing protein and electrolyte losses in wound exudate, preventing bacterial contamination, permitting less painful dressing changes, and improving the healing quality. This review overviews recent progress of collagen dressings for chronic skin wound healing. Various commonly used wound dressings for wound management have been first introduced. Collagen wound dressings have been categorized as films, sponges, hydrogels, nanofibers, and powders, and their efficacy has been compared. The critical functions of collagen dressings in wound healing, such as stopping bleeding, shortening inflammation, promoting angiogenesis, and stimulating tissue regeneration have been elaborated. The clinical applications of collagen dressings to repair different types of chronic wounds have been thoroughly summarized. A comprehensive list of commercialized collagen dressings has been updated, and an outlook of collagen dressings have been finally speculated. Graphical Abstract

The main objective of the present research study was to fabricate Vildagliptin floating microspheres by ionotropic gelation using natural polymer, Abelmoschus esculentus obtained from the fruits of Abelmoschus esculentus in combination with sodium alginate. Microspheres were prepared and optimized using central composite rotatable design model using design expert software version 12 .The study is focussed on the interaction effects of the three independent variables, natural polymer concentration, sodium alginate concentration and crosslinker concentration, optimization of formulations response surface methodology was used . drug –excipient compatibility studies were carried out by infrared spectroscopic studies. Infrared spectroscopic studies clearly shown that drug and excipients were compatible. Totally 15 formulations were generated taking 8 factorial points, 6 axial points and 1 centre point. Response surface methodology was used to optimize the formulations. To investigate the responses %cumulative drug release, floating time and floating lag time Response surface methodology was used Polynomial equations and model plots of 3 dimensional model surface plots were generated. Vildagliptin optimized microspheres were formulated and a second order, model quadratic model was used to study influence of formulation factors on response variables. Experimental data of Statistical analysis exhibited good coefficient of regression for cumulative in vitro drug release Regression F –ratios for the experimental variables were significant. The experimental values and predicted values are agreed. All fifteen formulations exhibited % yield of 94.35-99.99%, particle size of 124-441µm, %swelling index 64.52-89.65%, floating time of 10.16 to 13.16 and floating lag time of 30.31 to 46.98 sec. F4 formulation is optimized based on cumulative % in-vitro drug release at 2nd hour, 12th hour, floating lag time and floating time values. Predicted and observed results are in agreement 95% confidence intervals. Based on investigation, RSM is the good tool for optimization of formulations.

Sustained-release gastroretentive floating matrix tablets of cephradine were prepared for better patient compliance. Eight different tablets were prepared by using two natural polymers, psyllium husk powder (F1–F4) and xanthan gum (F5–F8), through the wet granulation technique. These tablets were characterized by pre- and postcompression analysis, Fourier transform infrared spectroscopy, swelling index study, in vitro buoyancy and dissolution study. Data were analyzed by model-dependent and model-independent analysis to devise the release mechanisms. The polymers exhibited excellent sustained-release behavior as well as binding characteristics. Pre- and postcompression parameters were observed in the specified official pharmacopoeia range. The drug contents of all the formulations were found in the range 95·52–99·63%. No chemical interaction was found between the drug and polymer. All formulations exhibited a good floating time – that is, >24 h – except F8, which remained buoyant for less than 1 h in simulated gastric fluid (pH 1·2). All of the formulations exhibited a direct relation between the swelling index and viscosity of polymer matrices. The significance of the wet granulation technique was indicated by the polymer action as a binding agent in wetting solution. From comparison of the two polymers, psyllium husk powder efficiently retarded the drug release owing to its high gelatinous swollen mass.
