A supersensitive immunofluorometric assay for rat luteinizing hormone

Abstract: We have developed an immunofluorometric assay (IFMA) for rat (r) LH, which is based on two monoclonal antibodies, one to bovine and the other to human LH. Signal detection occurs by time-resolved fluorescence evoked by a europium label (Delfia, Wallac). The method is fast in comparison to the standard RIA with the NIDDK reagents (4 h vs. 3 days). The sensitivity of the IFMA assay (0.75 pg/tube; NIDDK rLH RP-2) is over 30-fold higher than that of the NIDDK RIA (usual detection limit, 20-30 pg/tube). Using 25-mi… Show more

“…We used the ultra-sensitive DELFIA rat LH assay (Wallac Oy, Turku, Finland) to measure LH in rodent serum (27). Sensitivity was 0.03 ng/mL, and the intra-and interassay coefficients of variations at 33.1 ng/mL were 3.4% and 16.0%, respectively.…”

“…For seed enrichment analysis, only genes with FPKM above 1.0 were considered. For cumulative distribution function calculations, log 2 fold-change values were corrected for 3â€Č UTR length biases (55).…”

“…For intratesticular testosterone analysis, lipid extraction was performed using 2× diethylether. Serum LH and FSH concentrations were measured using immunofluorometric assays as described previously (55). For measuring intratesticular and serum testosterone, both sera and tissue homogenates were extracted twice with 2 ml of diethyl ether, followed by conventional RIA (56).…”

“…Serum levels of LH were measured by immunofluorometric assay (Delfia; PerkinElmer-Wallac, Turku, Finland) as described previously (Haavisto et al, 1993). The approximate assay sensitivity for LH was 0.0075 mg/l.…”
