A Systematic Review Exploring the Anticancer Activity and Mechanisms of Glucomannan

Abstract: Glucomannan, long recognized as the active ingredient of the traditional Chinese medicinal herb Konjac glucomannan, is a naturally occurring polysaccharide existing in certain plant species and fungi. Due to its special property to also serve as a dietary supplement, glucomannan has been widely applied in clinic to lower body weight and circulation cholesterol level and to treat constipation, diabetes, and arterial sclerosis. Besides the regulatory role engaged with gastroenterological and metabolic syndrome, … Show more

“…Mannose is a minor component of the EPSs produced by LAB, whereas in those produced by yeasts, it represents a major component. To date, different authors have reported the occurrence of a positive correlation between the high mannose content of EPS and their anticancer activity [83].…”

“…Because of the anti-inflammatory and antitumor effect of glucomannan, we used this compound for the first time to treat breast cancer-bearing mice as an adjuvant in the tumor lysate cancer vaccine model and as a dietary supplement [ 19 , 46 ]. Furthermore, using an autologous tumor lysate as a vaccine antigen was considered reliable against tumor development because it might consist of all the essential epitopes that could boost CD4+ assistant T cells and CD8+ T cells [ 47 , 48 ].…”

“…Today, glucomannan is usually associated with nutrition, regulating blood–lipid reduction, and suppressing diabetes and arterial sclerosis [ 17 , 18 ]. Studies have shown the suppression effect of glucomannan on colon, gastric, and hepatic cancers [ 19 , 20 , 21 ]. Chen et al discovered that glucomannan prevented the development of gastric cancer cell lines SGC-7901 and AGS in vitro.…”

“…Among them, nano-capsule based technology gains increasing attention. Many materials such as polyethylene glycol, polysaccharide, chitosan and liposome were used as nano-capsules to efficiently encapsulate the purified bioactive cargo for drug delivery through intravenous injection or oral administration ( Kolate et al, 2014 ; Li et al, 2019 ). Altogether, tremendous efforts are needed to improve the solubility and selectivity of aloperine for future clinical application.…”
