A thermodynamic study on the interaction between magnesium ion and human growth hormone

Abstract: A thermodynamic study on the interaction between magnesium ion and human growth hormone (hGH) was studied at 27 degrees C in NaCl solution (50 mM) using different techniques. Two techniques of ionmetry using a Mg2+selective membrane electrode and isothermal titration calorimetry were applied to obtain the binding isotherm for hGHMg2+; results obtained by both techniques were found to be in good agreement. There is a set of three identical and noninteracting binding sites for magnesium ions. The intrinsic disso… Show more

“…A thermodynamic study through ITC was performed which is a direct methodology of interpreting various thermodynamic parameters in various bio molecular interaction studies [37][38][39]. A reversed protocol was used throughout the experiment as the alkaloid has high heat of dilution [ Figure 5].…”

“…The calorimetric data analysis using equation (16) is a graphical fitting model, which extensively has been applied to studies on the enzyme inhibition [32][33][34]50] and metal binding to proteins [8,[58][59][60][78][79]. These two proposed data analysis for isothermal titration calorimetry is expected useful for a set of binding sites, especially for enzyme inhibition study and measurement of inhibition constant.…”

“…If α is defined as the fraction of free binding sites on the biomacromolecule, M 0 is the total biomacromolecule concentration and L 0 is the total ligand concentration, then the free concentrations of monovalent molecule, [M], and ligand, [L], as well as the concentration of bound ligand, [ML], can be deduced as follow [56,58]:…”

“…ITC gives invaluable information about biomacromolecule-ligand interactions. During the last two years we attempted to study the metal ion binding chemistry of different proteins [4][5][6][7][8][9][10][11][12]. They will change the conformational stability and form aggregates.…”
