Declined oocyte quality is the major contributor to female subfertility in aged mammals. Currently, there are no effective interventions to ameliorate aged oocyte quality. We found that oocytes from aged mice exhibited lower levels of mevalonate (MVA) pathway metabolites, including farnesyl pyrophosphate (FPP) and geranylgeranyl pyrophosphate (GGPP) and reduced cortical F-actin. We further demonstrated that MVA supplementation improved the FPP level, the cortical F-actin and the quality of aged oocytes. Mechanistically, we found that MVA supplementation induced granulosa cells to synthesize FPP, which was subsequently transferred to aged oocytes. Transported FPP increased small GTPases prenylation, including CDC42 and RAC1, and promoted membrane localization of CDC42-N-WASP-Arp2/3 and RAC1-WAVE2-Arp2/3 complexes, promoting cortical F-actin re-assembly and reducing aneuploidy of aged oocytes. We also identified an oral drug 8-isopentenyl flavone, as an isoprenoid donor from Epimedium brevicornu Maxim, which could increase CDC42 and RAC1 prenylation, improving the cortical F-actin and the competence of aged oocytes, ameliorating reproductive outcomes in aged female mice. Collectively, increasing small GTPases prenylation via MVA metabolites or 8-IPF provide a therapeutic approach for boosting fertility in women of advanced maternal age.

Diisobutyl phthalate (DiBP) is commonly used in the plastics industry, and recent studies have shown that environmental exposure and accumulation in the food chain caused inflammation in some organs. However, the underlying mechanisms by which DiBP affects oocyte quality have not yet been fully defined. We used immunostaining and fluorescence to evaluate the effects of DiBP exposure and demonstrated that it impaired the morphology of matured porcine oocytes through generation of cytoplasmic fragmentation, accompanied by the perturbed dynamics of the spindle and actin cytoskeleton, misdistributed endoplasmic reticulum, as well as partial exocytosis of cortical granules and ovastacin. Moreover, analysis of Smart RNA-seq found that DiBP-induced aberrant oocyte maturation could be induced by abnormal mitochondrial function and apoptosis. Importantly, we discovered that supplementation with pyrroloquinoline quinone (PQQ) significantly attenuated the meiotic abnormalities induced by DiBP exposure through the modulation of reactive oxygen species levels. Our findings demonstrated that DiBP exposure adversely affects oocyte meiotic maturation and that PQQ supplementation was an effective strategy to protect oocyte quality against DiBP exposure.

Hydrodynamic Mechanism for Stable Spindle Positioning in Meiosis II Oocytes

Cytoplasmic streaming, the persistent flow of fluid inside a cell, induces intracellular transport, which plays a key role in fundamental biological processes. In meiosis II mouse oocytes (developing egg cells) awaiting fertilization, the spindle, which is the protein structure responsible for dividing genetic material in a cell, must maintain its position near the cell cortex (the thin actin network bound to the cell membrane) for many hours. However, the cytoplasmic streaming that accompanies this stable positioning would intuitively appear to destabilize the spindle position. Here, through a combination of numerical and analytical modeling, we reveal a hydrodynamic mechanism for stable spindle positioning beneath the cortical cap. We show that this stability depends critically on the spindle size and the active driving from the cortex and demonstrate that stable spindle positioning can result purely from a hydrodynamic suction force exerted on the spindle by the cytoplasmic flow. Our findings show that local fluid dynamic forces can be sufficient to stabilize the spindle, explaining robustness against perturbations not only perpendicular but also parallel to the cortex. Our results shed light on the importance of cytoplasmic streaming in mammalian meiosis. Published by the American Physical Society 2024
