The prostate gland is the only male accessory gland in dogs and is responsible for secreting the prostatic fluid. Morphologically, the canine prostate gland lacks differentiation into zones, presenting a uniform parenchyma along the longitudinal axis. The luminal epithelial cells secrete a liquid rich in calcium, citric acid, simple sugars, and different enzymes as a component of the seminal plasma. Since the prostatic diseases are very common in small animal practice, there are many information regarding mechanisms of the different prostatic conditions and lack of information regarding the anatomy, histology, and physiology of the canine prostate gland. Thus, this chapter aims to meticulously describe the anatomy, histology, and physiology of the canine prostate gland.

The dog (canis lupus familiaris) is the only other species besides humans that develop spontaneous prostatic carcinomas (PCa) at a high frequency. The canine model is primarily utilized for the study of the PCa molecular mechanisms and provides a natural animal model for the study of potential therapies. In humans, the PCa frequently exhibits mutations in the C-MYC and a reduced expression of the E-cadherin and NKX3.1 proteins. This study's objective was to evaluate the NKX3.1, C-MYC, and E-cadherin expression in the canine normal prostate, benign prostatic hyperplasia (BPH), proliferative inflammatory atrophy (PIA) and PCa and to verify differences in expression and subcellular localization of these proteins in the prostatic carcinogenesis. A tissue microarray (TMA) slide was constructed, and immunohistochemistry with antibodies raised against C-MYC, NKX3.1, E-cadherin and p63 was performed using the peroxidase and DAB methods. The C-MYC protein expression was elevated in the cytoplasm and nuclei of the canine PCa and PIA compared with the normal prostate (P = 0.004. The NKX3.1 protein expression was reduced in 94.75% of the PCa and 100% of the PIA compared with the normal prostate (P = 0.0022). In fact, the expression of E-cadherin trended towards a decrease in carcinomas when compared to normal prostate and PIA. By immunohistochemistry, more p63-positive basal cells were observed in the PCa and PIA when compared with the normal prostate (P = 0.0002). This study has demonstrated that the carcinogenesis of canine prostatic tissue may be related to basal cell proliferation, the gain of C-MYC function and the loss of NKX3.1 protein expression.

Association of CD44+/CD24-Cells to More Aggressive Molecular Phenotypes in Canine Mammary Carcinomas

Leading the cause of death among female dogs, mammary gland tumor awakes the interest of the scientific community, mostly because the similarities in clinical presentation and evolution to breast cancer in women. Due to the importance of investigating canine mammary tumors, this research aims to identify the protein expression of CD44, CD24, MUC-1, CD10 and ESA by immuno-histochemistry. Thus, slides were prepared by the tissue microarray technique to assess the presence of these markers in different subtypes. Of the 112 tumors, 66.9% were positive for CD10 (75/112), 42.86% positive for CD24 (48/112), 21.43% for ESA (24/112), 20.54% for MUC-1 (23/112) and 5.35% for CD44 (6/112). Of these tumors, 3.57% showed the immuno-phenotype CD44+/CD24-(4/112). The results suggest that mammary gland tumors of female dogs were positives for stem cell markers, which may be associated with cancer stem cells. This association also may allow setting further models to study breast cancer in women.
