Abyssinone V‐4’ Methyl Ether Isolated from Erythrina droogmansiana (Leguminosae) Inhibits Cell Growth and Mammary Glands Hyperplasia Induced in Swiss Mice by the 7,12‐Dimethylbenz(a)anthracene

Abstract: There is a long standing interest in the identification of medicinal plants and derived natural products for developing cancer therapeutics. The present study was designed to evaluate the in vitro and in vivo antiproliferative effects of Abyssinone V-4' methyl ether (AVME) on breast tissue of mice. The cytotoxicity of AVME was evaluated using MTT assay in four cancer cell lines (DU145, PC3, HepG2, and MCF-7). Further, a protective effect of AVME was evaluated on 7,12-dimethylbenz(a)anthracene- (DMBA-) induced … Show more

“…e exposition to environmental pollutants such as DMBA induced mutation, DNA damage, oncogene activation, and cancer genesis [40,41]. e administration of a single dose of DMBA (50-80 mg/kgBW) to female rats was reported to induce breast cancer development [21,[41][42][43][44]. In agreement with this report, our results showed that, after the subcutaneous administration of a single dose of DMBA (50 mg/kgBW) under the mammary gland of female Wistar rats, breast tumors developed 22-26 weeks later, with an induction percentage of 40%.…”

“…Existing therapies are limited due to substantial adverse effects; therefore, search for novel agents with less or no side effects is warranted in this area. Recently, great interest has been raised in developing natural molecules as potential anticancer agents [18, 19]. In this view, various parts of the neem tree ( Azadirachta indica ) have been shown to inhibit the growth of cancer cells through several mechanisms [20].…”

“…The isolation of AVME has been previously reported by Tueche et al [ 23 ]. Briefly, 100 g of the ethyl acetate extract was subjected to column chromatography over silica gel packed in n-hexane.…”

“…Abyssinone V-4′ methyl ether (AVME, Table 1 ) also possesses estrogenic and antiestrogenic effects [ 15 , 22 ]. Recently, Tueche et al [ 23 ] reported the cytotoxic effect of AVME isolated from Erythrina droogmansiana on four tumoral cell lines [including estrogen receptor-positive breast adenocarcinoma (MCF-7)] and its ability to prevent breast tumors induced by 7,12-dimethylbenz(a)anthracene (DMBA) in mice. Given its aforementioned antiestrogenic and cytotoxic effects, AVME might be a good candidate for the treatment of estrogen-dependent cancers, mainly breast cancer.…”
