Acinetobacter calcoaceticus–baumannii Complex Strains Induce Caspase-Dependent and Caspase-Independent Death of Human Epithelial Cells

Abstract: We investigated interactions of human isolates of Acinetobacter calcoaceticus–baumannii complex strains with epithelial cells. The results showed that bacterial contact with the cells as well as adhesion and invasion were required for induction of cytotoxicity. The infected cells revealed hallmarks of apoptosis characterized by cell shrinking, condensed chromatin, and internucleosomal fragmentation of nuclear DNA. The highest apoptotic index was observed for 4 of 10 A.calcoaceticus and 4 of 7 A. baumannii stra… Show more

“…Acinetobacter spp. are known to invade epithelial cells and induce caspase-dependent and caspase-independent apoptosis [65]. Uptake of apoptotic bodies and caspase-dependant DNA fragmentation is known to facilitate LGT between mammalian cells [66], including LGT of oncogenes [67].…”

“…Adhesion and invasion ability were performed in a quantitative assay using the gentamicin/lysostaphin protection method, with modifications (Krzymińska et al 2012a, b, 2015). For the analysis of bacterial adhesion, the infected HeLa cells were lysed using 0.1% Triton X-100.…”

“…Two-fold dilutions (from 1:2 to 1:512) of supernatants were prepared, in phosphate buffered saline (PBS, Biomed). They were added to the HeLa cell monolayer and incubated for 24 h. The effect of extracellular toxins was expressed as a cytotoxic index and assessed as the reciprocal of the highest dilution of the culture filtrates, which produced a cytopathic effect that was observed under an inverted microscope (Krzymińska et al 2012a, b). As a negative control, cells treated with non-pathogenic E. coli K12C600 filtrate were used.…”

“…Recently, Acinetobacter calcoaceticus-A. baumannii complex strains associated with apoptosis in human epithelial cells were shown to induce the formation of autophagic vacuoles, although neither the mechanism nor the bacterial factor involved in this process was explored (36).…”
