Activable Multi-Modal Nanoprobes for Imaging Diagnosis and Therapy of Tumors

Abstract: Malignant tumors have become one of the major causes of human death, but there remains a lack of effective methods for tiny tumor diagnosis, metastasis warning, clinical efficacy prediction, and effective treatment. In this context, localizing tiny tumors via imaging and non-invasively extracting molecular information related to tumor proliferation, invasion, metastasis, and drug resistance from the tumor microenvironment have become the most fundamental tasks faced by cancer researchers. Tumor-associated micr… Show more

“…The characteristics of the tumor microenvironment play a major role in determining the success of cancer immunotherapies. 58 , 59 It is known that solid tumors can progressively develop several specific pathways to evade immune clearance, including through a microenvironmental remodeling process known as “cancer immunoediting”. This process imposes selective pressures within the tumor microenvironment, promoting the aggravation of cancer.…”

“…To address this issue, the concept of stimulus-responsive nanomaterials has been proposed. Such nanomaterials can be activated to exhibit therapeutic efficacy only in the tumor tissues under the stimulation of specific tumor pathological hallmarks, while remaining "silent" in normal tissues to ensure biosafety features [25][26][27][28][29]. For example, in our previous studies, a type of nanoprobes have been developed, which can specifically target the triple-negative breast cancer, be triggered by the acidic microenvironment, and responsively release the Fe 3+ only in tumor tissues to realize a safer tumor treatment [30].…”

“…[ 3 ] The malignant behaviours of tumour cells have been demonstrated to be associated with a series of molecular events inside tumour cells or in the tumour microenvironment, [ 4 ] therefore, identifying the abnormal expression of malignant pathological molecules is not only related to diagnosis and prognosis, but also has significance for therapeutic administration of cancer. [ 5 ] In previous studies and clinically statistical analyses of cancer, some key characteristics that closely related to the malignancy of tumour have been demonstrated, that is, acidic tumour microenvironment, abnormal secretion of various proteases such as hyaluronidase (HAase) and matrix metalloproteinases (MMPs), excessive glutathione (GSH), hypoxia, and abnormal redox state etc. [ 6 ] Nevertheless, most solid tumours display considerable complexity in vivo, leading to both spatial and temporal heterogeneity, which cannot be simply mimicked in vitro.…”
