Activated endothelial cells induce a distinct type of astrocytic reactivity

Abstract: Reactive astrogliosis is a universal response of astrocytes to abnormal events and injuries. Studies have shown that proinflammatory microglia can polarize astrocytes (designated A1 astrocytes) toward a neurotoxic phenotype characterized by increased Complement Component 3 (C3) expression. It is still unclear if inflammatory stimuli from other cell types may also be capable of inducing a subset of C3+ neurotoxic astrocytes. Here, we show that a subtype of C3+ neurotoxic astrocytes is induced by activated endot… Show more

“…In addition to the peripheral hepatic EFA metabolism and consequent brain transport, astrocytes have been suggested to be the central neural system providers of PUFA [ 64 , 65 ]. Surprisingly, based on our preliminary in vitro studies, human brain endothelial cells may be able to produce themselves DHA.…”

“…Reactive microglia also induce capillary permeability by producing ROS, and promoting the migration of blood-borne inflammatory cells through the BBB [117, 118]. Thus, it is clear that preventing microglial and astrocytic stress and over-reactivity can also promote vascular health, and vice-versa [101], as we observe in our CAI-treated mice.…”

“…Microglia, upon prolonged insults, prompt to proinflammatory phenotype, resulting in morphological and biochemical changes, which perpetuate brain inflammatory responses [97,98], while hindering clearance and degradation of unwanted products. All these clearance processes may act in parallel or in synergy to preserve brain homeostasis [24], and all appear to be dependent on vascular fitness and functionality, glial cell health, or both [4,26,99]. In his study, we show that the treatment with CAIs improves both cerebrovascular and glial fitness.…”

“…These findings build on a recent report that astrocytes activated by LPS‐treated endothelial cells—to which astrocytes in our model are exposed—exhibit a robust shift in their ECM remodeling profile. [ 42 ] Johnson et al. (2022) also recently found that, of 44 protein modules (families of related proteins) studied, enrichment in the matrisome protein module (collection of ECM‐associated proteins) was one of four modules most significantly correlated with AD neuropathology in both preclinical and clinical AD.…”
