“…In the current study, we found that p53 not only promoted Slug transcription, but also increased the levels of histone acetyltransferases p300 and CBP, and resulted in increased histone H3 and H4 acetylation. These epigenetic changes have been reported to regulate the terminal differentiation of osteoblastic progenitor cells and calcification of pAVICs (17)(18)(19)36). We suggest that p53 induced Slug expression initiates osteoblastic transdifferentiation at the early stage, and the epigenetic changes may contribute to the calcium deposition at the late stage of pAVIC calcification.…”

“…5G, H). p53 promotes pAVIC calcification by increasing slug transcription Since epigenetic changes of histone acetylation and associated histone acetyltransferase p300 and CREB-binding protein (CBP) have been reported promoting osteoblastic maturation and AVIC calcification (17)(18)(19). To dissect the mechanism of p53-regulated pAVIC calcification, we analyzed the epigenetic changes induced by p53.…”

“…As known, HP is capable of inducing epigenetic modifications, including DNA methylation and histone acetylation 23,39 , which mediate the effects of HP, such as fibrotic fibroblast activation and osteogenic transdifferentiation 39,40 . As cardiac hypertrophy and HF could be rescued by inhibition of DNA methylation or histone acetylation 41,42 , methylation and acetylation may also have important roles in the pathogenesis of cardiac hypertrophy.…”

“…Therefore, the aberrant expression of CREBBP may have impact on calcification which is consistent with the results of the present study. Furthermore, it was reported that the translocation of CiTED1 is enhanced by the protein kinase C activator (27,28). a number of studies have set out to determine the role of mir-330-3p in different types of tumors, most of which have reported mir-330-3p-3p as a tumor suppressor gene.…”

“…a number of classic alteration patterns are involved in calcification, including receptor translocation mediated by serine mutation and epigenetic modulation regulated by histone acetyltransferase (19,26). Previous studies reported the association between creBBP and aortic calcification, and suggested that CREBBP ultimately promotes calcification via its translocation from the cytoplasm to the nucleus (27). Taken together, the results of the present study and findings from previous studies indicated that the alteration of creBBP via mir-330-3p might aggravate susceptibility toward early calcification observed in patients with BAV.…”
