Eggs are perishable and fragile foods because of their easy contamination by different microorganisms, such as fungi. Hence, research on egg safety and quality has been recently developed worldwide to minimize illness transmitted by this food, although most present mainly pathogenic bacteria. Thus, this study identified potentially pathogenic fungal species in eggshells from free-range and commercial hens offered in supermarkets in Fortaleza, CE, Brazil. The research was performed at the Vettings laboratory at the State University of Ceará (UECE) with 40 eggs: 20 from commercial and 20 from free-range hens. The eggshells were assessed using the dextrose evaluation method for pathogenic cultivation, commensal, and yeast fungi. Subsequently, readings were taken to identify and count the fungi on a binocular light microscope in 100x increments. Both egg categories showed a significant number of colony-forming units (CFU). The study identified eight different fungal species in both egg production types. The most prevalent fungal colonies in the free-range hen lineage (brown shell) were yeast spp. and Penicillium citrinum, and the laying hen lineage (white shell) had mostly Penicillium spp. and yeast spp. Therefore, both egg production systems showed similar fungal contamination, which suggests that the manipulation and/or storage are adequate despite the lack of strict food safety regulations or supervision for eggs in Brazil.

We report a patient with severe cavitary pulmonary tuberculosis and Aspergillus niger superinfection, whose only comorbidity was untreated diabetes mellitus. A. niger pneumonia was proven by PCR, sequencing and culture of pleural and respiratory secretions. The patient was successfully treated with a four-drug antituberculous regimen, liposomal amphotericin B (up to 5 mg/kg/d) and pleuro-pneumonectomy. Histology of the resected lung revealed destroyed lung tissue with inflammatory cells and fungal conidia. There were large deposits of polarising material, which was found to be calcium oxalate. There was also nodular caseating necrosis bordered by epitheloid cells and connective tissue. Thus, all diagnostic criteria for invasive A. niger infection were met. Several local risk factors, such as extensive lung damage and tissue acidification, may have favoured superinfection by A. niger. This case highlights the diagnostic value of calcium oxalate crystals in lung tissue and the need for combined antimicrobial and surgical treatment in extensive invasive aspergillosis caused by A. niger.

Wprowadzenie i cel pracy. Aspergillus niger jest grzybem pleśniowym odgrywającym ważną rolę w rolnictwie i przemyśle spożywczym jako czynnik zakaźny roślin i zanieczyszczenie żywności. Szczepy A. niger mogą uwalniać mykotoksyny zagrażające zdrowiu bądź stanowić przyczynę poważnej choroby ludzi -aspergilozy. Leczenie infekcji grzybiczych wiąże się ze stosowaniem azoli, jednak coraz częściej obserwuje się lekooporność. Rozwiązaniem przełamania lekooporności mogą stać się olejki eteryczne cytrusów, które wykazują silne działanie przeciwdrobnoustrojowe. Celem pracy było ukazanie połączenia działania flukonazolu oraz olejku eterycznego pomarańczy Citrus aurantium oraz ocena charakteru interakcji farmakodynamicznej pomiędzy nimi przeciwko Aspergillus niger w badaniach in vitro. Materiał i metody. Doświadczenia prowadzono metodą hodowli płytkowych, oceniając strefy zahamowania wzrostu Aspergillus niger pod wpływem różnych dawek flukonazolu, olejku eterycznego pomarańczowego oraz kombinacji związków. Analiza izobolograficzna pozwoliła ocenić charakter interakcji pomiędzy testowanymi związkami. Wyniki. Testowane związki hamują wzrost A. niger w sposób zależny od stężenia. Wartość IC 50 flukonazolu względem A. niger wynosi IC 50 = 2,02 ± 0,79 mg/ml, natomiast olejku eterycznego pomarańczowego IC 50 = 3,78 ± 0,48%. Analiza izobolograficzna wykazała, że efektem kombinacji olejku eterycznego pomarańczowego i flukonazolu w stałej proporcji dawek 1:1 jest interakcja addytywna z tendencją do synergizmu w testach hodowlanych dla Aspergillus niger. Wnioski. Olejki eteryczne są naturalnymi związkami o wielkim potencjale terapeutycznym, zaś analiza izobograficzna może przyczynić się do wprowadzenia substancji naturalnych do terapii zakażeń lekoopornych.
