Acyl-CoA binding proteins; structural and functional conservation over 2000 MYA

Abstract: Besides serving as essential substrates for beta-oxidation and synthesis of triacylglycerols and more complex lipids like sphingolipids and sterol esters, long-chain fatty acyl-CoA esters are increasingly being recognized as important regulators of enzyme activities and gene transcription. Acyl-CoA binding protein, ACBP, has been proposed to play a pivotal role in the intracellular trafficking and utilization of long-chain fatty acyl-CoA esters. Depletion of acyl-CoA binding protein in yeast results in aberran… Show more

“…Cytosolic low-molecular mass (approximately 10 kD) acyl-coenzyme A-binding protein (ACBP) consists of a four-a-helix domain capable of binding acyl-CoAs with high affinity in a wide range of eukaryotic organisms (Faergeman et al, 2007). It is believed to serve a housekeeping function of maintaining free acyl-CoA concentrations at low nanomolar levels and, thus, prevents micelle formation and the partitioning of acyl-CoA into membranes (Knudsen et al, 1999).…”

“…It is highly conserved throughout evolution and expressed in all cell types in the eukaryotes investigated (1,2). This, together with the characteristics of the ACBP promoter (3,4), implies a housekeeping function of the gene.…”

“…In rats fasting for 24 hours significantly decreased tissue levels of ACBP in the liver, whereas feeding of a high-fat diet for 48 hours caused ACBP levels to increase [18]. In rainbow trout, the grouth rate of domesticated species were higher than wild species, ACBP expressinon changed in muscle and liver, the genes of domesticated fish mainly for the regulation and control, and a strong ability to control cell division and regulation of structural genes [19].…”

“…Acyl-CoA binding protein (ACBP) was known as diazepam binding inhibitor(DBI)/endozepine(EP), is a highly conserved, approximately 10 kDa cytosolic protein [1]. ACBP binds saturated and unsaturated C14-C22 acyl-CoA esters in a one-to-one binding mode with high specificity and affinity(Kd=2^10 nm) [2].…”
