ADAMTS proteases: key roles in atherosclerosis?

Abstract: The ADAMTS (a disintegrin and metalloproteinase with thrombospondin motifs) proteases are secreted enzymes that regulate extracellular matrix turnover by degrading specific matrix components. Roles for the proteases in inflammation and atherosclerosis have been suggested by a number of recent studies, and the role of ADAMTS-4 and -5 in the breakdown of aggrecan and subsequent degradation of cartilage during osteoarthritis has also been established. The ability of the ADAMTS proteases to degrade versican, the p… Show more

“…Proteases from the ADAMTS family regulate ECM turnover in atherosclerotic plaque [15], and hyaluronic acid (encoded by HAS2 gene) is a prominent constituent of the ECM in atherosclerotic vascular lesions [16]. Our analysis found several SNPs clustered near the ADAMTS5 gene (rs6816344, rs1452093, rs9977499, rs1817114, rs2830881) and a single SNP located near the HAS2 gene (rs17232789) to have significant correlations with survival.…”

“…ADAMTS family are circulating plasma enzymes responsible for processing procollagens and von Willebrand factor, and involved in cleavage of proteoglycans. 17,18) Numerous studies have shown that ADAMTS were key participants in the pathogenesis of vascular diseases, including atherosclerosis, aneurismal change, and restenosis. [19][20][21] ADAMTS proteases contributed to the plaque destabilization by weakening the fibrous cap, and expressed higher in coronary atherosclerotic plaques of ACS patients than SA patients.…”
