Adaptation of adhesion test using Caco-2 cells for anaerobic bacteriumPseudobutyrivibrio xylanivorans, a probiotic candidate

Abstract: Pseudobutyrivibrio xylanivorans strain Mz5(T), an anaerobic bacterium (originating from the rumen of a Holstein-Friesian cow), has some attributes that make it a possible probiotic strain (very active hydrolases, bacteriocin and conjugated linoleic acid production). For the estimation of its adhesion ability, the adhesion test on Caco-2 cells was introduced and adapted. The adhesion was performed in an anaerobic glove box in standard 24-well plates at neutral pH for 30 min. The best method for separation of th… Show more

“…Of those, 18 genera showed higher abundance in one rat genotype versus the other. Of the nine genera with higher abundance in BB-DR, three genera, Bifidobacterium, Lactobacillus and Pseudobutyrivibrio , have representatives with known probiotic activity (Silva et al , 1987; Vandenbergh 1993; Jack et al , 1995; Bernet-Camard et al , 1997; Lorca et al , 2001; Reid et al , 2003; Leahy et al , 2005; Ohkawara et al , 2005; Fukuda et al , 2006; Ohkawara et al , 2006; Salminen et al , 2006; Čepeljnik et al , 2003, 2007). These observations from pyrosequencing were verified by qPCR of Bifidobacterium and Lactobacillus .…”

“…However literature overview indicates that although viable probiotics have more documented health effects than nonviable probiotics, the latter are not without effect (Ouwehand and Salminen 1998;Čepeljnik et al 2007). Therefore, we proposed to evaluate the adhesion of fecal lactobacilli to cultured Caco-2 cells.…”
