Adipose-Derived Exosomes as Possible Players in the Development of Insulin Resistance

Abstract: Adipose tissue (AT) is an endocrine organ involved in the management of energy metabolism via secretion of adipokines, hormones, and recently described secretory microvesicles, i.e., exosomes. Exosomes are rich in possible biologically active factors such as proteins, lipids, and RNA. The secretory function of adipose tissue is affected by pathological processes. One of the most important of these is obesity, which triggers adipose tissue inflammation and adversely affects the release of beneficial adipokines.… Show more

“…sEV-AT also contributed to bone and soft tissue regeneration in the rat model of bisphosphonate-related osteonecrosis of the jaw [ 27 ]. In addition, due to the complex composition of adipose tissue-derived EVs, they may provide many opportunities to modulate the metabolism of distant tissues through the delivery of EVs, for instance, their regulation on endothelial cell vascularization via HIF-1α, Akt, ERK, and SDF-1 [ 35 ]. In the meantime, adipose tissue extract (ATE), the secretome of adipose tissue, has also been studied.…”

“…Nutritional stress, especially excessive dietary fats, leads to obesity and insulin resistance. This process is believed to be the result of impaired insulin signal transduction caused by several molecules released from adipose tissue [ 40 , 41 ]. However, not all fatty acids have a negative effect on insulin sensitivity [ 42 ].…”

“… 48 , 49 The released exosomes interact with target cells in three ways: (1) fusion of the plasma membrane between target cells and exosomes, (2) the binding of target cell membrane receptors with proteins on the membrane surface of exosomes, and (3) the in vivo transformation of exosomes by target cell endocytosis. 50 The origin and biogenesis of exosomes are mediated by tumour susceptibility gene 101 (TSG101), apoptosis-linked gene 2-interacting protein X (ALIX), endosomal sorting complexes required for transport (ESCRT) protein, soluble N-ethylmaleimide-sensitive factor (NSF) attachment protein receptor (SNARE) complex proteins, and other related substances. However, the specific mechanism underlying the role and function of these related substances in exosome biogenesis requires further investigation.…”
