Adipose Tissue - Adequate, Accessible Regenerative Material

Abstract: The potential use of stem cell based therapies for the repair and regeneration of various tissues offers a paradigm shift that may provide alternative therapeutic solutions for a number of diseases. The use of either embryonic stem cells (ESCs) or induced pluripotent stem cells in clinical situations is limited due to cell regulations and to technical and ethical considerations involved in genetic manipulation of human ESCs, even though these cells are highly beneficial. Mesenchymal stem cells seen to be an id… Show more

“…AT is a rich source of MSC with ubiquitous availability. Since large numbers of MSC can be obtained from AT by minimal invasive procedures, adMSC have become an important candidates for autologous and allogeneic stem cell based therapies and tissue engineering .Conversely, perinatal tissues such as the amnion, chorion and UC are promising MSC sources due to the young donor age . Although MSCs have been derived from multiple perinatal tissues, UC tissue (Wharton's jelly, WJ) and umbilical cord blood (UCB) are the most frequently used sources, with UC tissue being a richer MSC source compared to UCB .…”

“…A population of multipotent progenitors, the adipose-derived stem/stromal cells (ASCs) (long identified as preadipocytes) were identified by various studies including ours to exhibit such abilities (95,204,205,262,275,352). ASCs, as their bone marrow counterpart the mesenchymal stem/stromal cells (MSCs) are endowed with multilineage mesodermal differentiation potentials as well as regenerative abilities, leading to their extensive investigation from a therapeutic and tissue engineering perspective (77, 96,158). Adipose tissue remodeling is frequently reported to be associated with the infiltration of various cell populations (226,329).…”

“…The yield of ASC in the stromal vascular fraction of a lipoaspirate can reach 1 to 5%; this percentage differs depending on the harvesting site (Jurgens et al, 2008;Kolaparthy et al, 2015). For comparison, the isolation of bone marrow mesenchymal stem cells requires a painful procedure, and a relatively small percentage of stem cells is present in the bone marrow aspirate (500 times less than ASC; Mizuno, 2009).…”
