Adipose tissue can be generated in vitro by using adipocytes from human fat tissue mesenchymal stem cells seeded and cultured on fibrin gel sheet

Abstract: The current study has developed an innovative procedure to generate ex novo fat tissue by culturing adipocytes from human fat tissue mesenchymal stem cells (hFTMSCs) on fibrin gel sheet towards applications in medicine and cosmetology. Fibrin gel has been obtained by combining two components fibrinogen and thrombin collected by human peripheral blood. By this procedure it was possible to generate blocks of fibrin gel containing adipocytes within the gel that show similar features and consistency to human fat t… Show more

“…In order to have a long-term stable biocompatible interface and keep the manufacturing process straightforward, we chose to modify PLLC surface with fibrin coating because fibrin is an insoluble and fibrous protein formed from blood fibrinogen polymerization and it has been developed in varieties of biomedicine applications. 25,28,29 This well established surface modification achieved scaffold's superior biocompatibility. Yet, it can further enhance tensile strength of the porous materials designed to be readily degradable.…”

“…The hADSCs were from an available cell line and were isolated, cultured, and expanded according to the protocol described previously (Tran et al, 2013). hADSCs were determined to positively express CD13, CD44, CD73, CD90, CD105, and CD166, and they differentiated into adipocytes after culturing for 21 days in adipogenic medium (Tran et al, 2013). In this study, hADSCs at the 4th passage were used.…”

“…Besides this group, some other groups in Vietnam have also been involved in stem cell research. One stem cell group, headed by Dr. Tran Cong Toai, at Pham Ngoc Thach Medicine University (HCMC), has investigated procedures to collect and expand stem cells in vitro from bone marrow and adipose tissues [50][51][52]. This group has also conducted studies on culturing and differentiating MSCs on scaffold in order to produce bone [53].…”
