Advances in Aptamers

Abstract: Aptamers are nucleic acid sequences synthesized through in vitro selection and amplification technique, possessing a broader range of applications in therapeutics, biosensing, diagnostics, and research. Aptamers offer a number of advantages over their antibodies counterpart, one of them is their ability to undergo chemical derivatization to increase their life in the body fluids and bioavailability in animals. Although aptamers were discovered in 1990s, they have become one of the most widely investigated mole… Show more

“…They are also stable during long-term storage and easy to transport at an ambient temperature. Similar to antibodies, aptamers can also be labeled with reporter molecules such as fluorescein, biotin, and digoxigenin, which increases their applicability for further applications (39)(40)(41)(42)(43)(44)(45). DNA and RNA aptamers have shown similar function and performance in terms of affinity and specificity (46).…”

“…Incorporation of modified nucleotides during SELEX requires the use of mutant T7 polymerases that will utilize these NTPs by in vitro transcription. These modifications minimize degradation of RNA oligonucleotides by nucleases (Bunka, Platonova et al 2010;Syed and Pervaiz 2010).…”

“…One aptamer is already being used as a therapeutic agent The small size of aptamers is advantageous for accessing their targets in vivo, but this also results in rapid renal clearance. To extend retention time in the system, aptamers are conjugated with PEG to increase molecular weight (Syed and Pervaiz 2010).…”
