Naturally occurring polysaccharides and/or their chemically modified derivatives have been widely investigated in relation to their use as components of controlled release systems for drug delivery. The aforementioned is due, in part, to their distinct properties such as abundant availability and biocompatibility as well as environmental and economic advantages. Chitosan (CS) and xanthan gum (XG) based matrices have received growing scientific/pharmaceutical interest as oral controlled release drug carriers. Herein, recent advances spanning the last two decades in CS-XG based drug delivery systems are reviewed with the emphasis being on oral tablet formulations, due to their versatility as pharmaceutical dosage forms. The mechanism of interaction between CS and XG, by means of computational and experimental approaches, is scrutinized. Results obtained from the literature establish the possibility of fabricating a controlled release drug delivery system based on CS and XG matrices. This can be achieved by monitoring and manipulating the physiochemical properties of the two polymers as well as the experimental variables affecting their drug retardation efficiency, without the need to employ special equipment or sophisticated experimental techniques/methodologies.

Microfabrication technology has enabled the development of novel controlled-release devices that possess an integration of structural, mechanical, and perhaps electronic features, which may address challenges associated with conventional delivery systems. In this feature article, microfabricated devices are described in terms of materials, mechanical design, working principles, and fabrication methods, all of which are key features for production of multifunctional, highly effective drug delivery systems. In addition, the current status and future prospects of different types of microfabricated devices for controlled drug delivery are summarized and analyzed with an emphasis on various routes of administration including ocular, oral, transdermal, and implantable systems. It is likely that microfabrication technology will continue to offer new, alternative solutions to design advanced and sophisticated drug delivery devices that promise to significantly improve medical care.

BACKGROUND Most drugs are designed for immediate release via oral administration, releasing the dosage rapidly. Those administration forms can present a wide variation in drug concentrations, which can be harmful to a patient, when the amount of drug is either below or above the therapeutic range. Modified‐release formulations promise to alleviate the problems inherent to immediate‐release drugs by modifying their form of encapsulation. A sericin/alginate blend was applied as a matrix to modify the release of furosemide. The influence of furosemide and alginate concentration was evaluated in the incorporation efficiency and release time of 85% of drug content using a 22 full‐factorial experimental design. Various analytical techniques were employed to characterize the produced drug particles. RESULTS The incorporation efficiencies ranged from 73.4 ± 2.1% to 84.7 ± 2.9% and the release could be classified as delayed and sustained due to gastroresistance of the particles and a longer release time compared to the commercial drug. Release mechanism was controlled by the relaxation of macromolecular chains due to absorption of water by the particles and the erosion of the matrix. The presence of furosemide crystals was confirmed. Furosemide thermal stability was not altered with its incorporation into the sericin/alginate blend and mean diameter was small enough that the particles could be applied to multiparticulate systems. CONCLUSIONS Sericin/alginate blend could serve as a biodegradable and biocompatible matrix for furosemide incorporation for delayed and sustained drug delivery. © 2020 Society of Chemical Industry (SCI)
