Affimer proteins as a tool to modulate fibrinolysis, stabilize the blood clot, and reduce bleeding complications

Abstract: Bleeding complications secondary to surgery, trauma, or coagulation disorders are important causes of morbidity and mortality. Although fibrin sealants are considered to minimize blood loss, this is not widely adopted because of its high cost and/or risk for infection. We present a novel methodology employing nonantibody fibrinogen-binding proteins, termed Affimers, to stabilize fibrin networks with the potential to control excessive bleeding. Two fibrinogen-specific Affimer proteins, F5 and G2, were identifie… Show more

“…As described in this report, we established a versatile tool, POLArIS, taking advantage of Adhiron/affimer that can be screened through phage display for any specific target molecules of interest. There are many Adhirons/affimers reported, targeting a variety of specific biomolecules (19,20,(42)(43)(44)(45)(46)(47)(48) from laboratories, and the production service is commercially available. The two-axis fluorescence polarization imaging is a robust system for mapping fluorescence anisotropy with the aid of commonly used software for image analysis (NIS-Elements Advanced Research from Nikon and ImageJ).…”

“…A newer approach to target PECAM-1 or G6b-B would be affimers; non-antibody binding proteins derived from a scaffold engineered from human stefin A and a cystatin consensus sequence [88]. While affimers are in the early stages of development, promising results have been accomplished stabilizing fibrin networks with potential reduction on bleeding risk, with a fibrinogen-binding affimer [89]. Activatory affimers targeting G6b-B/PECAM-1 would be promising therapy, although this has yet to be attempted.…”

“…The structure of the Escherichia coli immunoglobulin binding proteins (Eibs) showed how they are involved in entero–hemorrhagic pathogenicity [ 94 , 95 ]. Another innovative tool that was developed for use in molecular recognition applications was the use of “adhirons” to help stabilize complexes and to gain structural information (e.g., [ 96 ]). Adhirons are non-antibody scaffold binding proteins [ 97 ].…”
