Albomycin Uptake via a Ferric Hydroxamate Transport System ofStreptococcus pneumoniaeR6

Abstract: The antibiotic albomycin is highly effective against Streptococcus pneumoniae, with an MIC of 10 ng/ml. The reason for the high efficacy was studied by measuring the uptake of albomycin into S. pneumoniae. Albomycin was transported via the system that transports the ferric hydroxamates ferrichrome and ferrioxamine B. These two ferric hydroxamates antagonized the growth inhibition by albomycin and salmycin. Cross-inhibition of the structurally different ferric hydroxamates to both antibiotics can be explained b… Show more

“…Transport across the cytoplasmic membrane is mediated by the ABC transporter FhuBCD (37). Upon release of the active antibiotic moiety (aminoacyl-thioribosyl pyrimidine) from the iron-chelating group (40), it interferes with the loading of serine to the seryl-tRNA by seryl-tRNA synthetases (41). Interestingly, we found that the Npp permease of P. aeruginosa is also able to translocate albomycin when heterologously expressed in E. coli.…”

“…Albomycins are composed of a tris(N 5 -acetyl, N 5 -hydroxyornithine) peptide and a nucleoside-analogous thioribosyl pyrimidine moiety linked by a serine spacer (17). Because of its similarity to fungal ferrichromes, albomycin is recognized by the FhuA receptor and the FhuD binding protein, which was shown by both mutagenesis (259) and crystallization studies (58). After hydrolysis from the siderophore carrier, the serylthioribosyl pyrimidine moiety blocks protein biosynthesis by inhibiting seryl-tRNA synthetases (307).…”

“…S2). Thus, the protein would facilitate iron exchange, which is first order in both Fe-and apo-siderophore, by increasing the local concentration of the apo-siderophore (15 (27). In Gram-negative bacteria not only A. hydrophila (14) but also E. coli (28) and Salmonella typhimurium enterica (29) have OMTs for FO/Fch import.…”
