Albumin depletion method for improved plasma glycoprotein analysis by two-dimensional difference gel electrophoresis

Abstract: Figure 3. Tryptic peptides found in α-1-antichymotrypsin. The amino acid sequence of α-1-antichymotrypsin is shown. The sequence is divided into discrete tryptic peptides, which are color-coded according to the legend at the bottom of the figure. The color coding signifies whether the peptide is found in the mass spectrum shown in Figure 2.

“…Hence, failure to correct for variable Tg content may introduce a severe bias to protein expression studies of human thyroid tissues. A likewise problematic scenario has long been noted for albumin in serum samples (39)(40)(41). While there are kits claiming to deplete albumin from serum samples, there is none for human thyroglobulin.…”

“…Affinity chromatography is usually used for purifying the target protein with high affinity binding upon the matrix used; for example, a molecule of antibody or enzyme substrate directed interact a specific protein is attached to the bead. Otherwise, the lectin-agarose affinity chromatography based on lectin specificity on carbohydrate moiety is also used to bind specific sugars for depletion of carbohydrate moieties or enrichment of glycoproteins prior to proteomic analysis [36]. Gel filtration chromatography is used to separate proteins or peptides on the basis of size.…”

“…These include use of the affinity dye ligand, Cibacron blue, binding to immobilized lectin, or absorbing the serum sample with immobilized antibody specific for human albumin [41][42][43][44]. None of these procedures have proved very efficient and all are expensive.…”
