All-Optical Modulation of Single Defects in Nanodiamonds: Revealing Rotational and Translational Motions in Cell Traction Force Fields

Abstract: Measuring the mechanical interplay between cells and their surrounding microenvironment is vital in cell biology and disease diagnosis. Most current methods can only capture the translational motion of fiduciary markers in the deformed matrix, but their rotational motions are normally ignored. Here, by utilizing single nitrogen-vacancy (NV) centers in nanodiamonds (NDs) as fluorescent markers, we propose a linear polarization modulation (LPM) method to monitor in-plane rotational and translational motions of t… Show more

“…However, despite the huge potential of existing diamond-based PUF labels 38,39,48 , it has not yet been possible to achieve the much-desired high-dimensional (> 2D) encoding. We here propose a method to achieve a 3D encoded diamond-based PUF label, based on the linear polarization modulation (LPM) 47 curves of FNDs with random orientations.…”

“…Practically, the localized measurement of nanoscale physical quantities such as magnetic fluctuations, electric signals, and temperatures inside or across the membrane has been demonstrated at the early stage. Besides the outstanding spin properties, researchers have also started to harness other optical properties to capture the rotational motions of single NV centers in NDs [ 170 ]. This orientation sensing capability of NV centers can be utilized to detect the membrane force and traction to a subcellular level, which undoubtedly will lead to a new path for a deeper understanding of membrane behaviors and motions.…”
