Allelic variability in the third intron of the fibroin light chain gene in Bombyx mori (Lepidoptera: Bombycidae)

Abstract: ABSTRACT. Conformation-sensitive gel electrophoresis is a useful method for identifying allele polymorphism; it provides co-dominant molecular markers. Using this method, we identified genetic variability in the third intron of the fibroin light chain gene, fib-L, in six Bombyx mori strains. Only Chinese C21A strain did not demonstrate allelic alterations, showing only homoduplex DNA molecules. We found distinct heteroduplex profiles in the Japanese HAA, M12B and M19-2 and the Chinese C25B and C24-2 strains. A… Show more

“…A pool of first-instar larvae from both strains (M11A and C24A) were macerated with liquid nitrogen, and DNA was extracted as previously described (Barbosa et al, 2008(Barbosa et al, , 2009. DNA fragments were amplified using the primers described in the DNA sample section.…”

“…DNA fragments were amplified using the primers described in the DNA sample section. As a control, the third intron of the B. mori light fibroin chain gene was amplified from C122B and C25B strains, as described elsewhere (Barbosa et al, 2009). The C122B strain is negative for the heteroduplex genotype (Barbosa JB, personal communication), while the C25B strain presents a positive result for the CSGE technique (Barbosa et al, 2009).…”

“…FIBH has one intron and two large exons, while Fib-l gene has seven exons separated by large introns. The latter gene mostly contains non-coding DNA as the first intron represents 60% of the gene, and the rest of the introns add up to 31% (Barbosa et al, 2009).…”
