Allergen Screening Bioassays: Recent Developments in Lab-On-A-Chip and Lab-On-A-Disc Systems

Abstract: Allergies occur when a person's immune system mounts an abnormal response with or without IgE to a normally harmless substance called an allergen. The standard skin-prick test introduces suspected allergens into the skin with lancets in order to trigger allergic reactions. This test is annoying and sometimes life threatening. New tools such as lab-on-a-chip and lab-on-a-disc, which rely on microfabrication, are designed for allergy testing. These systems provide benefits such as short analysis times, enhanced … Show more

“…Some affinity separation techniques reviewed above are ready to be integrated with microfluidic system, such as magnetic beads. Though there are not yet reports on such system designed for active compounds screening, it is possible in theory due to the common procedures of incubation, wash, and elution that already existed [71]. Lab-on-a-chip or lab-on-a-CD is another feasible strategy for high-throughput screening, which would contribute significantly to chemical and pharmacological research of TCM.…”

“…Flow control is achieved by varying the spinning speed, spinning direction, and operations such as valve opening, siphoning, volume metering, mixing, and flow switching. In our previous studies, a LOAD platform was built for allergy testing [ 16 , 17 ]. In our second generation LOAD, the AO-loaded cells that were sensitized with a patient’s serum or medium alone were challenged with suspected allergens or other triggers in chamber at body temperature (37 °C).…”

“…One obvious point of development is towards low-cost, portable and disposable point of care diagnostic (POC) devices. [1][2][3][4] In virtually all POC implementations, there is a need for a multifunction matrix material onto which operations are affected. Many polymeric materials such as polyethylene, nitrocellulose (NC), Dacron, polyvinyl chloride (PVC), nylon, polyacrylonitrile (PAN) etc.…”
