Alphavirus polymerase and RNA replication

Abstract: Alphaviruses are typically arthropod-borne, and many are important pathogens such as chikungunya virus. Alphaviruses encode four nonstructural proteins (nsP1-4), initially produced as a polyprotein P1234. nsP4 is the core RNA-dependent RNA polymerase but all four nsPs are required for RNA synthesis. The early replication complex (RC) formed by the polyprotein P123 and nsP4 synthesizes minus RNA strands, and the late RC composed of fully processed nsP1-nsP4 is responsible for the production of genomic and subge… Show more

“…nsP2 has helicase, ATPase, GTPase, and RNA 5′-triphosphatase as well as protease activities. nsP4 is the RNA-dependent RNA polymerase with a conserved C-terminal GDD RNA polymerase motif and adenylyl transferase (20,21). Identification of the function(s) of nsP3, a phosphoprotein found both in replication complexes and cytoplasmic aggregates (22,23), has been more difficult.…”

“…This is the first step in extending our knowledge of alphavirus membrane-associated RCs, and further studies on these purified complexes should give insights into the host proteins present and required for their activity on the plasma membrane, as well as their structural organization. Currently, alphaviruses, coronaviruses, and arteriviruses represent important groups of pathogenic positivestrand RNA viruses lacking structural information on RdRps (57,58). The alphaviral core RdRp has been purified in an active form (31), but the structure of the RC might give more information about the active conformation of the polymerase and interactions, as all four nsPs are required for RNA synthesis.…”

“…Replication of their genomes is accomplished either by RNA-dependent RNA synthesis ( Fig. 2A, B) [reviewed in (Ferron et al, 2017;Lu and Gong, 2017;Menéndez-Arias and Andino, 2017;Pietilä et al, 2017;Tao and Ye, 2010)] or through RNAdependent DNA synthesis (reverse transcription) followed by DNA integration, replication, and transcription ( Fig. 2C).…”
