Anxiety disorder is a psychiatric condition reported in diabetic patients. It is known that hypothalamus-pituitary-adrenal axis activity is increased in these patients and corticosteroids levels are augmented, whereas the anxiolytic actions of diazepam are reduced. The aim of this study was to evaluate the participation of glucocorticoid (GR) and mineralocorticoid (MR) receptors in anxiety in diabetic mice and whether the blockade of these receptors synergizes with diazepam in the diabetic condition, leading to a reduction of anxiety. Diabetes was induced with streptozotocin (STZ) and anxiety-like levels were evaluated on days 5, 15, and 30 after STZ. Independent groups of control and diabetic mice were treated with diazepam (0.25-2.0 mg/kg), RU-486 (12.5-100 mg/kg), spironolactone (12.5-100 mg/kg), or the combination of suboptimal doses of diazepam and MR or GR antagonists. Results showed that STZ increased anxiety-like behavior 15 days after its administration. The response to diazepam was reduced in diabetic mice, whereas GR and MR blockade induced anxiolytic-like effects in these animals. Coadministration of MR or GR antagonists synergized with diazepam to induce anxiolytic-like effects. The results suggest the participation of corticosteroid receptors in the increased anxiety-like response in diabetic mice and that the blockade of these receptors facilitates the effects of diazepam.

Abstract Background and Objectives: Obesity has reached epidemic proportions globally. Among several methods for treating obesity, the use of dietary supplements is common recently. One supplement that can help in this regard might be vitamin B6 in high doses.The objective of this study was to evaluate the efficacy of pyridoxine hydrochloride supplementation on anthropometric measurements, body composition, visceral adiposity index and metabolic status in obese and overweight women.Methods and Study Design: In this randomized controlled clinical trial, 44 obese and overweight women aged 18–50 years were selected and divided randomly into two groups: an intervention group (receiving 80 mg pyridoxine hydrochloride supplement for 8 weeks) and control group (receiving placebo for 8 weeks). Results: In the pyridoxine hydrochloride group, weight(p=0.01), body mass index(p=0.009), fat mass(p=0.003), waist circumference(p=0.005), visceral adiposity index(p=0.001), fasting insulin, Insulin Resistance(HOMA-IR), Total cholesterol, low density lipoprotein(LDL), Triglycerides (TG) and leptin(p<0.001) decreased whereas adiponectin(p<0.001) increased in comparison to the baseline values. There was a significant difference in fat mass, visceral adiposity index, fasting insulin, HOMA-IR and TG between pyridoxine hydrochloride and control groups following intervention in adjusted models(p<0.05).Conclusions: The findings suggest that vitamin B6 supplementation may be effective in reducing BMI and improving body composition and biochemical factors associated with obesity.Trial registration: The trial was approved by the Ethics Committee of Ahvaz Jundishapur University of Medical Sciences (reference number: ir.ajums.rec.1394.254) and was registered at Iranian Registry of Clinical Trials (code: IRCT20181002041206N1).

Когнітивна дисфункція в дітей: можливості фармакологічної корекції із застосуванням комплексу амінокислот та вітаміну В 6 Резюме. До когнітивних функцій належить здатність розуміти, пізнавати, вивчати, усвідомлювати, сприймати і переробляти зовнішню інформацію. Серед основних причин виникнення когнітивної дисфункції виділяють перинатальне ушкодження центральної нервової системи або асфіксію, екологічні фактори. При когнітивній дисфункції в дітей спостерігаються порушення розвитку навчальних навичок, моторних функцій, змішані специфічні розлади розвитку. Найчастіша скарга -зниження концентрації уваги відмічається у 21 % дітей України. Когнітивна дисфункція часто поєднується з депресією, тривожністю, адитивною поведінкою; практично в половини пацієнтів переходить у дорослий вік. Серед механізмів розвитку когнітивної дисфункції ключову роль відіграють оксидативний стрес та низькоінтенсивне запалення в нейронах. За результатами експериментальних та клінічних досліджень доведено, що компоненти комплексного препарату Когівіс амінокислоти аспарагін і глутамін, а також DL-фосфосерин і вітамін В 6 сприяють розвитку мозку; забезпечують синтез білків, нуклеотидів, нейротрансмітерів, глутатіону тощо; впливають на оксидативний стрес у центральній нервовій системі, зокрема в гіпокампі. Все це створює умови для оптимального функціонування нейронів і розвитку когнітивних функцій.
