Alterations in mandibular morphology associated with glypican 1 and glypican 3 gene mutations

Abstract: Alterations in GPC3 expression are likely to mediate changes to mandibular size in craniosynostosis. These findings have potential future applications in the prevention and treatment of craniosynostosis and associated craniofacial dysmorphology.

“…Micro‐computed tomography (micro‐CT) imaging has been a powerful tool in studies of craniofacial phenomics , root canal morphology and mineral density of dental hard tissues . Continuing development and availability of micro‐CT imaging provide additional avenues to explore changes to the dental tissues both in health and disease , enabling the varying aetiological and clinical characteristics of the various dental anomalies to be explored.…”

“…Furthermore, SDC1 has been reported as a unique regulator of bone-cell communication, as osteoclasts lacking SDC1 have diminished differentiation and bone resorption potential ( Kim et al, 2018 ; Timmen et al, 2020 ). GPC3 is an important bone co-regulator, and GPC3-deficient mice resulted in reduced mandibular growth and decreased osteoclast differentiation ( Viviano et al, 2005 ; Mian et al, 2017 ). Cadherins are cell adhesion molecules, which are homologous to calcium-dependent glycoproteins.…”

“…Traditional morphometric analysis of the murine craniofacial complex is based on linear measurements, which can be performed using most commercial 3D image processing software packages such as Mimics, Avizo, and CTAn (see Note 15). Here, we describe acquisition of standard linear measurements as described previously [6][7][8][9].…”

“…Micro-CT is highly suitable for traditional morphometric analysis of the craniofacial complex in small animals. This includes analyses using 3D landmark-based linear distances, surface areas, and volumes to characterize animal phenotypes in health and disease states [6][7][8][9][10][11]. Micro-CT can be also used to complement histological studies, allowing nondestructive analysis of gross morphology alongside histological investigations.…”
