Altered DNA methylation in estrogen-responsive repetitive sequences of spermatozoa of infertile men with shortened anogenital distance

Abstract: Background It has been suggested that antenatal exposure to environmental endocrine disruptors is responsible for adverse trends in male reproductive health, including male infertility, impaired semen quality, cryptorchidism and testicular cancer, a condition known as testicular dysgenesis syndrome. Anogenital distance (AGD) is an anthropomorphic measure of antenatal exposure to endocrine disruptors, with higher exposure levels leading to shortened AGD. We hypothesized that exposure to endocrin… Show more

“…A negative effect of estrogen was found in the fractions of spermatozoa from infertile men with shortened ano-genital distance. This fraction is more likely to contain transposable elements harboring an estrogen receptor response element and their sperm shows substantial hypomethylation in estrogenic Alu sequences [ 40 ]. In summary, our observations are consistent with a postulated role of estrogen in modulating the expression of enzymes that modify histones, impacting the histone code.…”

“…This variation has been suggested to represent variation in protamine levels between sperm 17 and has been correlated with poor reproductive outcomes 16 . To determine if NicE-view high sperm and CMA3 high sperm represent the same sub-population, we used FACS (Fluoresecence Activated Cell Sorting) of sperm stained with CMA3 to obtain CMA3 low and CMA3 high enriched sperm populations 28 . We then performed our T4 NicE-view assay on these two populations (isolated from 6 individuals).…”

“…Samples used for FACS isolation of sperm based on Chromomycin A 3 (CMA3) levels were obtained from participants from an additional cohort (recruitment of which was also approved by the Ethikkommission Nordwest-und Zentralschweiz (EKNZ 2017-01407)) and whose sperm were previously examined for alterations in DNA methylation 28 . This cohort consisted of a control group of fertile individuals who had previously donated sperm (and whose semen parameters were within the WHO standard ranges) and a group of infertile individuals with normal semen parameters, but anogenital distance (AGD) of less than 40 mm 29 and >20% TUNEL positive swim-up sperm in a previous donation.…”
