Amperometric Biosensor Based on Diamine Oxidase/Platinum Nanoparticles/Graphene/Chitosan Modified Screen-Printed Carbon Electrode for Histamine Detection

Abstract: This work describes the development and optimization studies of a novel biosensor employed in the detection and quantification of histamine in freshwater fish samples. The proposed biosensor is based on a modified carbon screen-printed electrode with diamineoxidase, graphene and platinum nanoparticles, which detects the hydrogen peroxide formed by the chemical process biocatalysed by the enzyme diamine oxidase and immobilized onto the nanostructurated surface of the receptor element. The amperometric measureme… Show more

“… Schematic diagrams of the reactions involved in the quantification of biogenic amines . (A) Quantification of histamine based on hydrogen peroxide formation and either direct assessment of the latter in a platinum electrode (solid line, Apetrei and Apetrei, 2016 ) or through the use of horseradish peroxidase (dashed line, Pérez et al, 2013 ). (B) Quantification of tyrosine through oxidation with tyrosinase to dopaquinone and reduction of the latter to dopamine on the surface of a carbon electrode (Apetrei and Apetrei, 2015 ).…”

“…An excellent correlation between the histamine amount values obtained with biosensors and the histamine amount values obtained by ELISA (enzyme-linked immunosorbent assay ) and HPLC has been established [ 14 , 36 , 37 ]. Therefore, its use for screening purpose is suitable to analyze the sample enrichments and detect those contaminated with strong HPB active at low or ambient temperatures.…”

“…In addition to carbon based materials, NM of noble metals having size dependent fascinating optical, electro‐magnetic, and chemical properties have been comprehensively followed not only for their important technical contemplation, but also for their numerous industrial applications . NP of metals, particularly gold , silver, and platinum, are among the utmost comprehensively considered NM in the design of sensors for clinical diagnosis, bio‐imaging, and targeted drug delivery . It is due to their oxide‐free surface, ease of conjugation with biological entities, great compatibility with biological counterparts, and exceptional opto‐electronic properties .…”
