An efficient and comprehensive plant glycerolipids analysis approach based on high‐performance liquid chromatography–quadrupole time‐of‐flight mass spectrometer

Abstract: In past two decades, numerous lipidomics approaches based on mass spectrometry with or without liquid chromatography separation have been established for identification and quantification of lipids in plants. In this study, we developed an efficient and comprehensive lipidomics approach based on UPLC with an Acquity UPLCTM BEH C18 column coupled to TripleTOF using ESI in positive ion mode and MS/MSALL scan for data collection. Lipid extract was prepared to 2 mg/ml solution according to dry tissue weight and mi… Show more

“…Seed oil biosynthesis starts approximately 20 days after flowering (DAF), and oil accumulates actively at 40 DAF in B. napus (Lu et al, 2019b). Thus, we sequenced seed transcriptomes at two developmental stages (309 accessions at 20 DAF and 274 accessions at 40 DAF; Supplemental Table 4).…”

“…Accurate molecular weight and fragmentation ion information were acquired using LipidView 2.0 to generate an in-house dataset. The identification of glycerolipid classes and species was based on retention time, accurate m/z, and fragmentation ion patterns according to a previous report 50 .…”

“…Lipidomics analyzes membrane lipid composition and has been used to decipher the role of lipids on tolerance to abiotic stress conditions [56]. Electrospray ionization tandem mass spectrometry (ESI-MS/MS) has been traditionally used for lipidomics although other mass spectrometry instruments such as HILIC-ESI-IT-TOF-MS/MS or UPLC-TripleTOF make lipid analyses more efficient and accurate [57]. Although this is a promising approach to study plant stress, its use in roots is limited.…”
