An Electrochemical DNA Biosensor for the Detection of the Apa I Polymorphism in the Vitamin D Receptor Gene Using Meldola's Blue as a Hybridization Indicator

Abstract: Electrochemical detection of nucleic acid base mismatches related to Apa I single nucleotide polymorphism (SNP) in the vitamin D receptor gene was performed successfully using 7-dimethyl-amino-1,2-benzophenoxazinium salt (Meldolas blue, MDB) with 10.9 pmol/100 mL of detection limit. MDB reduction signals obtained from probe, mismatch(probe-SNP containing target) and hybrid(probe-target) modified pencil graphite electrode(PGE) increased respectively. The sensor was able to clearly distinguish perfect match from… Show more

“…Vyskyocil et al 196 used a DNA biosensor based on a screen printed CPE immobilized with double-stranded DNA for in detection of genotoxic nitro derivatives. Aladag et al 197 used DNA biosensors to detect Apa I polymorphism, which are involved in Alzhiemer’s Disease, within 30 minutes, which is more rapid than other current assays. Zhang et al 198 used DNA biosensors for simultaneous detections of both HIV-1 and HIV-2 oligonucleotides.…”

“…However, the reduction signal of MDB increases following the hybridization because MDB can selectively bind to duplex form of DNA. 13 In addition, when using PGA coated electrode, biosensor has shown superior electrocatalytic activity on MDB reduction signal (Figure 2).…”

“…Following hybridization, the DNA biosensors were immersed for 5 min in 20 mM tris buffer solution (TBS) (pH 7.0) containing 20 mM MDB. 12,13 Then, the biosensors were immersed in washing buffer (1×SSC+ 0.1% SDS, pH 7.0) for 5 min. The DNA sensor was then dipped into the TBS (pH 7.0) for voltammetric measurement.…”

“…These mutations can be used to trace generational inheritance patterns associated with specific diseases [55]. In addition, the developed biosensor compares favorably to other DNA biosensors based on a similar method for the hybridization detection (Table 2SI), considering important analytical properties, such as sensitivity and detection limit [51,43,[56][57][58][59].
