Leaf extracts from wild pepper (Capsicum flexuosum Sendt) were analysed for the presence of malate dehydrogenase (E.C. 1.1.1.37; MDH) isozymes using starch gel electrophoresis. Seven phenotypes for MDH isozymes were observed in the genitors. Genetic analysis in F 1 progenies revealed five loci coding for MDH. Isozyme banding patterns of hybrids indicated that MDH-3 and MDH-4 genes code for monomeric enzymes, while MDH-5 for a dimeric isoform. In MDH-2 loci, one particular F 1 progeny showed a significant deviation from the expected isozyme pattern. It is possible that other genes are controlling the expression of MDH-2 in pepper. Also, there are two alleles coding for MDH-2 isozyme. Sete fenótipos de malato desidrogenase foram observados entre os genitores. As análises de segregação em progênies F 1 revelaram que cinco locos gênicos estavam envolvidos na codif icação de MDH. Os padrões de bandeamento dos híbridos indicaram que os genes MDH-3 e MDH-4 codificavam para enzimas monoméricas, enquanto o MDH-5, para uma isoforma dimérica. Para o loco MDH-2, detectou--se desvio significativo para proporção de segregação esperada. Outros genes podem estar controlando a expressão de MDH-2 em pimenta. Como nos outros locos MDH, detectaram--se dois alelos codificando para MDH-2. ABSTRACT. Genetic diversity and structure were analyzed in 10 accessions belonging to Banco Ativo de Germoplasma de Capsicum located at Federal University of Piauí in northwestern Brazil that receives pepper samples grown in community gardens in various regions and Brazilian states. Selections were made from seeds of C. chinense (4 accessions), C. annuum (5 accessions), and C. baccatum (1 accession). Samples consisting of leaves were collected from 4-10 plants of each accession (a total of 85 plants). Native polyacrylamide gel electrophoresis was used to identify α-and β-esterase polymorphisms. Polymorphism was clearly detected in 5 loci. Sixteen alleles were found at 5 α/β-esterase loci of the three Capsicum species. In the C. chinense samples, the highest H O and H E values were 0.3625 and 0.4395, respectively, whereas in C. annuum samples, H O and H E values were 0.2980 and 0.3310, respectively; the estimated H O and H E values in C. chinense samples were higher than those detected in C. annuum samples. A deficit of homozygous individuals was found in C. chinense (F IS = -0.6978) and C. annuum (F IS = 0.7750). Genetic differentiation between C. chinense and C. annuum at these loci was high (F ST = 0.1867) indicating that C. chinense and C. annuum are genetically structured species for α/β-esterase isozymes. The esterase analysis showed high genetic diversity among the C. chinense and C. annuum samples and very high genetic differentiation (F ST = 0.6321) among the C. chinense and C. annuum samples and the C. baccatum accession.

Als meus i al llaurador valencià RESUMENLas hortalizas en general, entre ellas los pimientos, se encuentran continuamente expuestas al ataque de patógenos. No obstante, las enfermedades de etiología viral destacan por ser el principal factor limitante de estos cultivos. La estrategia más eficaz para el control de las virosis en plantas consiste en la identificación de genes de resistencia y su posterior utilización para el desarrollo de variedades resistentes. Una importante parte de la variabilidad existente en el género Capsicum se encuentra conservada en bancos de germoplasma. Sin embargo, para poder manejarla y explotarla eficientemente es necesario cuantificar y analizar la variabilidad interespecífica e intraespecífica disponible. Además, el disponer de una estrategia capaz de disminuir los costes asociados al proceso de fenotipado será muy ventajoso.El principal objetivo de esta tesis se centra en la caracterización y aprovechamiento de la variabilidad genética de las especies cultivadas del género Capsicum (C. annuum, C. chinense, C. frutescens, C. baccatum y C. pubescens). Así, en este trabajo se plantea tanto conocer y describir la distribución de la variabilidad existente en estas especies, como desarrollar herramientas que permitan una exploración y utilización eficiente de la variabilidad contenida en los bancos de germoplasma. Para ello, se ha puesto a punto una plataforma de EcoTILLING que permite un cribado eficiente de la colección de entradas de Capsicum del Banco de Germoplasma del COMAV. A modo de experiencia piloto se exploró esta plataforma para la búsqueda de variantes alélicas de los genes eIF4E y eIF(iso)4E, implicados en la resistencia a virosis de gran importancia en el cultivo del pimiento y de otros cultivos.Esta tesis doctoral es un ejemplo donde el EcoTILLING basado en el ADNc resulta fructífero, al trabajar con diferentes especies del género Capsicum y al ser los cambios nucleotídicos de las regiones codificantes los más interesantes.
