This chapter aims to provide an introduction to how engineering tools in general and computational models in particular can contribute to advancing the tissue engineering (TE) field. After a description of the current state of the art of TE, the developmental engineering paradigm is briefly discussed. Subsequently an overview is provided of different model categories that focus on different aspects of TE. These categories consists of the models that focus on either the TE product, the TE process or the in vivo results obtained after implantation. Generally, in all these models the aim is firstly to understand the biological process at hand and secondly to design strategies in silico to enhance the desired in vitro or in vivo behaviour. Finally, the need for quantification and parameter determination is discussed along with the computational tools and models that can be used to design the thereto required experiments in the most intelligent way.

An increasing number of novel and highly specialized computer-aided decision-making technologies for short-term production scheduling in oil refineries has emerged and evolved over the past two decades, thereby encouraging refiners to permanently rethink the way the refining business is operated and managed. In this report, we discuss the key lessons learned from one of the pioneering, yet daring, enterprise-wide programs entirely implemented in an energy company devoted to developing and implementing an advanced refinery production scheduling (RPS) technology, i.e., the RPS system of Petrobras. Apart from mathematical and information technology issues, the long-term sustainability of a successful RPS project is, we argue, the outcome of a virtuous cycle grounded on permanent actions devoted to improving technical education inside the organization, reinspecting organizational cultures and operational paradigms, and developing working processes.

Developmentally Engineered Callus Organoid Bioassemblies Exhibit Predictive In Vivo Long Bone Healing

Clinical translation of cell‐based products is hampered by their limited predictive in vivo performance. To overcome this hurdle, engineering strategies advocate to fabricate tissue products through processes that mimic development and regeneration, a strategy applicable for the healing of large bone defects, an unmet medical need. Natural fracture healing occurs through the formation of a cartilage intermediate, termed “soft callus,” which is transformed into bone following a process that recapitulates developmental events. The main contributors to the soft callus are cells derived from the periosteum, containing potent skeletal stem cells. Herein, cells derived from human periosteum are used for the scalable production of microspheroids that are differentiated into callus organoids. The organoids attain autonomy and exhibit the capacity to form ectopic bone microorgans in vivo. This potency is linked to specific gene signatures mimicking those found in developing and healing long bones. Furthermore, callus organoids spontaneously bioassemble in vitro into large engineered tissues able to heal murine critical‐sized long bone defects. The regenerated bone exhibits similar morphological properties to those of native tibia. These callus organoids can be viewed as a living “bio‐ink” allowing bottom‐up manufacturing of multimodular tissues with complex geometric features and inbuilt quality attributes.
